黄芩黄酮对自由基引起的大鼠脑线粒体损伤的保护作用

摘    要:目的 探讨黄芩中主要四种黄酮—黄芩素、黄芩苷、汉黄芩素和汉黄芩苷对脑线粒体氧化损伤的保护作用。方法 运用抗坏血酸 Fe2 +、2 ,2 azobis ( 2 amidinopropane)hy drochloride (AAPH)及NADPH三种不同损伤体系诱导大鼠大脑皮质线粒体脂质过氧化 ,研究黄芩中四种主要黄酮在不同体系中的保护作用。结果 四种黄酮在 10 μmol·L-1下对NADPH诱导的脂质过氧化都具有保护作用 ,而只有黄芩素和黄芩苷在前二体系中效果显著 ;四种黄酮对抗坏血酸 Fe2 +体系诱发的线粒体肿胀和膜的流动性降低都有一定的保护作用。结论 黄芩黄酮能抑制不同体系诱导的线粒体氧化损伤 ,黄芩素在各项测定中效果最好

Abstract:AIM To evalueate the protective effects of four main flavonoids in Scutellaria baicalensis Georgi on oxidative injury of rat cortex mitochondria METHODS Lipid peroxidation was induced by three different system, egascorbic acidFe 2+ , 2,2azobis (2amidinopropane) hydrochloride and NADPH; the swelling of mitochondria and the decreasing of membrane fluidity were induced by ascorbic acidFe 2+ system RESULTS Baicalein, baicalin, wogonin and wogonoside at 10 μmol·L -1 can protect mitochondria from NADPH induced lipid peroxidation, while only baicalein and baicalin showed remarkable effects on ascorbic acidFe 2+ and AAPH induced lipid peroxidation The swelling of mitochondria and the decreasing of membrane fluidity induced by ascorbic acidFe 2+ system can be retarded more or less by these four flavoids CONCLUSION Flavonoids in Scutellaria baicalensis Georgi may protect mitochondria from oxdative injury induced in different ways Baicalein is the most effective flavonoids among the tested agents in different systems
