氨基葡萄糖对绝经后骨质疏松模型大鼠的保护作用研究

摘    要:目的 探讨氨基葡萄糖对绝经后骨质疏松大鼠的作用效果及机制。方法 以小鼠颅顶前骨细胞MC3T3-e1为体外模型,给予不同浓度氨基葡萄糖,检测细胞增殖率,碱性磷酸酶含量,矿化结节数量;并通过剪除SD大鼠双侧卵巢的方法建立体内绝经后骨质疏松模型,以氨基葡萄糖(250、100 mg/kg)对模型大鼠进行连续12w的灌胃治疗,检测与骨相关的血清生化指标(ALP、Ca、P),三点弯曲试验检测大鼠股骨的生物力学性能,原子吸收法检测骨矿Ca含量,以及病理切片分析大鼠骨组织结构,从而研究氨基葡萄糖对骨质疏松大鼠的作用效果与机制。结果 氨基葡萄糖体外促进成骨细胞增殖,诱导成骨细胞的分化;并在体内逆转因切除卵巢后引起的骨量丢失、降低的骨强度和损伤的组织结构。结论 氨基葡萄糖对骨质疏松具有一定的改善作用。

Abstract:Objective To investigate the effects of D-glucosamine on postmenopausal osteoporosis. Methods The mice anterior parietal cells of MC3T3-e1 were selected as the in vitro model, and the proliferation rate, content of alkaline phosphatase and the number of mineralized nodules were detected. Female SD rats were ovariectomized and different doses of D-glucosamine were orally administered respectively, the sham and model group rats were intragastric administration therapy. Serum, uterus, femur and tibia tissues were made and preserved in certain condition to measure serum biochemical parameter, femoral mechanical properties, bone quality and bone tissue structure after oral administration for 3 months. Result D-glucosamine could significantly promote proliferation and differentiation of MC3T3-e1 cells. In addition, D-glucosamine could reverse ovariectomized-induced bone loss, deteriorative microarchitecture, decreased bone strength and impaired histologic structures of rats. Conclusion D-glucosamine has a positive effect on postmenopausal osteoporosis.
