摘    要:目的 对1株南海沉积物来源的PPtase高表达放线菌Streptomyces sp. SCSIO 40060进行次级代谢产物研究。方法 利用硅胶柱色谱、薄层色谱、Sephadex LH-20凝胶柱色谱和高效液相色谱(HPLC)等手段对PPtase高表达菌株发酵产物进行分离纯化;运用核磁共振(NMR)、质谱(MS)等波谱方法,并结合相关文献数据比对,鉴定化合物的结构。结果 从PPtase高表达菌株发酵产物中分离纯化得到7个化合物,包括5个二酮哌嗪类化合物、1个萘醌衍生物和1个萘甲酸衍生物,分别为methoxyneihumicin (1)、XR334 (2)、(S,Z)-3-benzylidene-6-isopropylpiperazine-2,5-dione (3)、(S,Z)-3-benzylidene-6-methylpiperazine-2,5-dione (4)、(3Z,6Z-3-(4-methoxybenzylidene)-6-(2-methylpropylidene)-piperazine-2,5-dione (5)、2-methoxy-1,4-naphthoquinone (6)、1-hydroxy-4-methoxy-2-naphthoic acid (7)。

Abstract:Objective To study the secondary metabolites from the PPtase overexpressed marine-derived actinomycete Streptomyces sp. SCSIO 40060 isolated from a South China Sea sediment sample. Methods The fermentation extract of the PPtase overexpressed strain was separated and purified by silica gel column chromatography, thin layer chromatography, Sephadex LH-20 gel column chromatography and high-performance liquid chromatography (HPLC). The structures of these compounds were determined on the analyzing of the NMR and MS spectroscopic data and comparisons with the data from reported literatures. Results Seven compounds, including five diketopiperazines, a derivative of naphthoquinone and a derivative of naphthoic acid, were isolated from the fermentation extract of the PPtase overexpressed strain and identified as methoxyneihumicin (1), XR334 (2), (S,Z)-3-benzylidene-6-isopropylpiperazine-2,5-dione (3), (S,Z)-3-benzylidene-6-methylpiperazine-2,5-dione (4), (3Z,6Z)-3-(4-methoxybenzylidene)-6-(2-methylpropylidene)-piperazine-2,5-dione (5), 2-methoxy-1,4-naphthoquinone (6), 1-hydroxy-4-methoxy-2-naphthoic acid (7).
