摘    要:目的研究养殖喘水苔海绵Tedania anhelans化学成分及其生物活性,为药源海绵规模化养殖和开发提供依据。方法综合利用薄层色谱、硅胶色谱、ODS C18色谱、Sephadex LH-20凝胶柱色谱、半制备高效液相色谱(HPLC)等方法对化学成分进行了分离;通过质量光谱测定(MS)、核磁共振(NMR)等方法并结合相关文献,对化合物结构进行鉴定。结果从养殖喘水苔海绵Tedania anhelans中分离得到13个化合物,分别为胆固醇(1)、3β-羟基-24-亚甲基胆甾-5-烯-7-酮(2)、胆甾-5-烯-3β,7α-二醇(3)、胆甾-5-烯-3β,7β-二醇(4)、3β-羟基胆甾-5-烯-7-酮(5)、胸苷(6)、去甲基胸苷(7)、环(脯氨酸-缬氨酸)二肽(8)、piperazirum(9)、(E)-4-(1H-indol-3-yl)but-3-en-2-one(10)、3-吲哚乙酸甲酯(11)、3-吲哚甲醛(12)、毛脉五味子醇(13)。结论13个化合物均为首次从该种海绵中分离得到,化合物8在50μmol·L?1浓度水平对HSV-2病毒抑制率为60.7%。

Abstract:Objective To study the chemical composition and biological activity of the cultured sponge Tedania anhelans,and provide a basis for the large-scale breeding and development of medicinal sponges.Methods Comprehensive use of thin layer chromatography,silica gel chromatography,ODS C18 chromatography,Sephadex LH-20 gel column chromatography,semi-preparative HPLC,and other methods to separate the chemical components;MS,NMR,and other methods combined with related literature,the structures of the compounds were identified.Results 13 compounds were isolated from the cultured sponge T.anhelans,namely cholesterol(1),3β-hydroxy-24-methylenecholest-5-en-7-one(2),and cholesta-5-ene-3β,7α-diol(3),cholesta-5-ene-3β,7β-diol(4),3β-hydroxycholesta-5-en-7-one(5),Thymidine(6),desmethylthymidine(7),Cyclo(prolinevaline)(8),Piperazirum(9),(E)-4-(1H-indol-3-yl)but-3-en-2-one(10),3-Indole methyl acetate(11),3-indole carboxaldehyde(12),pubinernoid A(13).Conclusion All 13 compounds were isolated from this sponge for the first time.Compound 8 showed an inhibition rate of 60.7%against the HSV-2 virus at a concentration level of 50μmol·L?1.
