南瓜多糖的分离、纯化及其降血脂作用

摘    要:目的 :研究南瓜多糖对正常及糖尿病模型小鼠血脂的影响。方法 :南瓜经甲醇回流、热水提取、弃蛋白、透析、乙醇沉淀 ,再经SephadexG1 0 0柱层析纯化 ,得南瓜多糖 (PP)。聚丙烯酰胺凝胶电泳证明其为均一体。PP的分子量为 1 .6× 1 0 4。经纸层析表明由D 葡萄糖、D 半乳糖、L 阿拉伯糖、木糖和D 葡萄糖醛酸组成。其摩尔比分别为 6∶3∶2∶1∶6。结果 :PP水溶液 2 0 0、5 0 0mg/kg经腹腔注入正常及糖尿病模型小鼠后 ,两者总胆固醇、低密度脂蛋白下降 ,高密度脂蛋白增加。结论 :PP是较理想的能改善脂类代谢的食疗剂。

Abstract:Purpose:In order to investigate the effect of pumpkin polysaccharide (PP) on normal mice and alloxan induced diabetic mice. Methods:From the pumpkin, a polysaccharids was extracted with hot water and purified by deproteinization and removal of lipid. Some chemical and physical properties of the polysaccharide were examined. It was homogeneous as judged by electrophoresis. PP molecular weight was estimated to be 1.6×10 4 and composed mainly of D glucose, D galactose,L arabinose, xylose and D glucuronicacid and in the molar ratios of 6∶3∶2∶1∶6 respectively. PP was given by the route of ip at a dose of 200mg/kg and 500mg/kg. Results:PP was capable to decrease blood lipid in normal mice, alloxan induced diabetic model mice, and showed dose effect relation to a certain extent. Conclusion:PP is a ideal practical food drug for treating lipid metabolism.
