盐效应分离纤溶活性化合物FGFC1

摘    要:目的探索通过盐效应分离长孢葡萄穗霉FG216(Stachybotrys longispora 216)发酵液中纤溶活性化合物FGFC1(fungi fibrinolytic compound 1)的方法。方法长孢葡萄穗霉FG216经过种子液培养和发酵培养获得的发酵液经浓缩后添加氯化钠、氯化铵和氯化钙至不同饱和度,用乙酸乙酯萃取,通过HPLC检测FGFC1的分离效果,并分析FGFC1的质谱和光谱特性。结果随着氯化钠、氯化铵和氯化钙的饱和度从20%升高到90%,氯化钠和氯化钙的盐效应分离作用效果呈现逐渐增强的趋势,60%饱和度的氯化钠能达到18.8 mg/mL的FGFC1提取率,是甲醇分离方法的118%,采用盐效应分离和用甲醇分离获得的FGFC1的质谱和光谱特性一致。结论用氯化钠盐效应分离长孢葡萄穗霉FG216发酵液中FGFC1是一种有效的分离方法,该盐效分离结果符合盐效分离的基本理论。

Abstract:Purpose To explore the isolation method of fibrinolytic compound FGFC1(fungi fibrinolytic compound 1) by salt effect from the broth of Stachybotrys longispora FG216.Methods The broth was concentrated after the salts including sodium chloride,ammonium chloride and calcium chloride were added,respectively,following that the concentrated liquor was extracted with ethyl acetate.The separation effect of FGFC1 was determined by HPLC,mass spectrometry and spectral characteristics.Results Experiment with the different saturations of sodium chloride,ammonium chloride and calcium chloride increased from 20% to 90%.The isolation effect of sodium chloride and calcium chloride showed a gradual increasing trend.When the saturation of sodium chloride was 60%,the extraction rate of FGFC1 can reach 18.8 mg/mL,which accounted for 118% of methanol extract.The mass and spectral characteristics of FGFC1 are consistent with those extracted by salt effect and extracted by methanol.Conclusion Isolating fibrinolytic compound FGFC1 by salt effect is an effective method of separation and the salt separation efficiency accords with the basic theory of salts effective separatian.
