摘    要:本研究采用Sephadex G-100柱层析,用中性Tris-HCl缓冲液洗脱,分离眼镜蛇全毒.结果不仅有效地分离出神经再生因子和神经毒素,而且保持了各自的生物活性.

Abstract:The cobra venom was dissovled in neutral Tris-HCl buffer and separated through Sephadex G-100 column (1. 5 × 90 cm) with the same buffer. Seven peaks were obtained. The nerve growth factor (NGF) was in peak Ⅱ and Ⅲ , while the neurotoxin (NT) was in peak Ⅵ. Mixed peak Ⅱ and Ⅲ solutions and chromatographied on CM-Cellulose 22 column (2. 0×40 cm), the NGF activity was eluted out in the eluent with 0. 4 mol/L NaCl. The isolated NGF had the activity of stimulating gangli-a growth at the concentration lower to 0. 05 μg/ml. The LD50 of NT on mice (18-22 g) was 3. 18 μg.
