摘    要:目的比较3种测定低分子量透明质酸(LMWHA)相对分子质量(Mr)方法。方法分别采用特性黏度法、高效凝胶渗透色谱(HPGPC)法及多角度激光散射仪结合凝胶渗透色谱(GPC/MALLS)法测定LMWHA的Mr,并对所得结果进行对比分析。结果对于Mr介于10 000~100 000范围内的LMWHA,特性黏度法测得的Mr值与GPC/MALLS法测得的Mr值基本相同,而HPGPC法由于所用标准Mr对照品结构上的差异,测得的Mr与其它两种方法测得的值差异较大。结论在一定Mr范围内,特性黏度法和GPC/MALLS法均可用于测定LMWHA的Mr。

Abstract:Purpose To compare the feasibility of three methods in determining the relative molecular weight(Mr) of low molecular weight hyaluronic acid(LMWHA).Methods Intrinsic viscosity method,high performence gel permeation chromatography(HPGPC) and multi-angle laser light scattering combined with gel permeation chromatography(GPC/MALLS) were used to determine the Mr of LMWHA,and the results were analyzed and compared with each other.Results In the Mr range of 10 000~100 000,the Mr values of LMWHAs obtained by intrinsic viscosity method were almost the same as those detemined by GPC/MALLS.Because of the structure difference between LMWHA and standard molecular weight polysaccharides,the Mr values obtained by HPGPC were significantly different from those obtained by the other two methods.Conclusion Within certain molecular weight range,both intrinsic viscosity method and GPC/MALLS can be used to determine the Mr of LMWHA.
