青黛提取物对HEL细胞增殖、凋亡和分化的影响

摘    要:目的研究青黛有效成分在体外对HEL细胞的增殖、凋亡和分化作用。方法应用MTT法检测青黛有效成分对HEL细胞的增殖抑制效应;流式细胞术检测CD14和CD11B,观察HEL细胞单核系分化趋势;流式细胞术AnnexinV-FITC/PI双标记法检测青黛有效成分对HEL细胞的早期凋亡诱导作用。结果与阴性对照组相比25,50,75和100μg/mL青黛有效成分作用下HEL细胞均出现单核系分化趋势,其中100μg/mL的分化趋势最明显;MTT结果显示25,50,75及100μg/mL药物对HEL细胞的生长抑制率分别为16.01%,39.12%,50.06%和59.28%;青黛有效成分可诱导HEL细胞凋亡:25,50,75及100μg/mL药物对HEL的早期凋亡率分别是5.60%,13.7%,17.8%和18.4%,明显高于对照细胞的自发凋亡率(2.70%)(P<0.05)。结论青黛有效成分对体外培养的HEL细胞具有显著抑制增殖作用,且可诱导细胞早期凋亡和单核系分化。
