山楂熊果酸的制备及对小鼠免疫功能和肝癌细胞凋亡的影响

摘    要:目的探讨山楂熊果酸(UA)对环磷酰胺(CTX)造成的免疫低下小鼠的保护作用及UA对HepS肝癌细胞的影响。方法腹腔注射CTX形成免疫抑制模型,观察UA对模型小鼠的免疫活性的影响;从小鼠腹腔抽取对数生长期肝癌细胞,用流式细胞术和DNA凝胶电泳法检测不同剂量UA对肿瘤细胞凋亡的影响。结果3个剂量的UA均能显著升高外周血的白细胞数,增强腹腔巨噬细胞的吞噬功能,能促进脾淋巴细胞增殖,增加脾指数;凝胶电泳检测DNA梯带,不同剂量的UA均能显著增加肿瘤细胞的凋亡率。结论UA对CTX造成的免疫抑制小鼠有显著的正调节作用,对HepS肝癌细胞凋亡有显著促进作用。

Abstract:Purpose To explore the protective effect of ursolic acid from hawthorn fruits to immunosuppressive mice caused by cyclophosphamide(CTX) and the effect of UA on hepatocellular carcinoma HepS cell.Methods Immunosuppressive model with CTX by ip was established.The influence of UA in immunological function on model mice was observed.The effect of UA on the cell apoptosis of HepS tumor cell was assessed by flow cytometrial analysis and DNA agarose gel electrophoresis.Results Every UA group could significantly increase the peripheral leucocyte count and enforce phagocytic activity of peritoneal macrophage.But as far as enhancing the proliferation of spleen lymphocyte and increasing spleen index as concerned,UA groups at four different dose levels can improve the rate of apoptosis.Conclusion UA displayed obviously positive regulation in immunosuppressive mice and improved the apoptosis of hepatocellular carcinoma HepS cell.
