高效液相色谱法同时测定人血清中氨茶碱、苯巴比妥、苯妥英钠及卡马西平的浓度

目的：建立同时测定人血清中氨茶碱（APL）、苯巴比妥（PB）、苯妥英钠（DPH）和卡马西平（CBZ）的高效液相色谱（HPLC）法。方法：以Agilent TC-C18（250 mm×4.6 mm,5μm）为分析柱,甲醇-水（60∶40）为流动相,检测波长230 nm,流速1.0 ml/min,柱温35℃。结果：APL、PB、DPH、CBZ均能达到良好的分离,与相邻峰的分离度（R）均〉1.5;线性范围分别为1.01～40.34 mg/L（r=0.999 5）、1.24～59.52 mg/L（r=0.999 1）、2.60～31.20 mg/L（r=0.998 6）、1.09～32.61 mg/L（r=0.999 2）;平均回收率分别为98.15%、99.25%、98.86%、100.3%;日内和日间精密度的RSD均〈4.3%。结论：本法灵敏、准确、简便、快速,适用于临床血药浓度的检测。

Simultaneous Determination of Aminophylline, Phenobarbital, Phenytoin Sodium and Carbamazepine in Serum by HPLC

OBJECTIVE：To establish an HPLC method for simultaneous determination of aminophylline （APL） , phenobarbital （PB）, phenytoin sodium （DPH） and carbamazepine （CBZ） in serum. METHODS： The samples were separated on Agilent TC-Cls （250 mm ×4.6 mm, 5 μm） analytical column. The mobile phase consisted of methanol- water （60： 40） with a flow rate of 1.0 ml/min. The detection wavelength was set at 230 nm and column temperature was set at 35 ℃. RESULTS： APL, PB, DPH and CBZ were well-separated （R 〉 1.5 for all）. The linear ranges of APL, PB, DPH and CBZ were 1.01-40. 34 mg/L（r =0. 999 5） ,1.24-59.52 mg/L（r =0. 999 1） ,2.60-31.20 mg/L （ r = 0. 998 6 ） and 1.09-32.61 mg/L （ r = 0. 999 2 ）, respeetively. The average reeovery rates were 98.15%, 99.25%, 98.86% and 100. 3%, respectively. Both the inter-day RSD and intra-day RSD were 〈 4.3%. CONCLUSION： This method is proved to be sensitive, aeeurate, simple, rapid and suitable for elinieal determination of serum drug concentration.
