摘    要:目的研究芹菜素(apigenin)诱导体外培养人成骨肉瘤MG-63细胞凋亡作用及其机制。方法体外培养MG-63细胞,分光光度法测定细胞内谷胱甘肽(GSH)含量;流式细胞术检测细胞凋亡率;琼脂糖凝胶电泳观察细胞DNA断裂。结果 apigenin显著降低MG-63细胞内GSH水平(P〈0.05),呈浓度依赖性。apigenin以浓度依赖方式诱导MG-63细胞凋亡率增高(P〈0.05),并导致细胞DNA断裂;500μmol/LGSH预孵育1h,能有效阻断apigenin诱导MG-63细胞内GSH耗竭和细胞凋亡作用(P〈0.05)。结论 apigenin通过耗竭MG-63细胞内GSH诱导成骨肉瘤细胞凋亡。

Abstract:Objective To investigate whether apigenin(API) induces apoptosis through glutathione (GSH) depletion in human osteogenic sarcoma MG-63 cell line. Method Human osteogenic sarcoma MG-63 cells were cultured in vitro. The intracellular level of GSH was measured using spectrophotometer. The percentage of apoptotic death cells was examined by flow cytometry (FCM) using propidium iodide staining. DNA fragmentation was examined using DNA agarose gel electrophoresis. Results Apigenin significantly reduced the intracellular level of GSH (P 0.05) in MG-63 cells, in a concentration-dependent manner. Apigenin significantly induced apoptosis of MG-63 cells(P 0.05), in a concentration-dependent manner. Pretreated with 500μmol/L GSH for 1 h can effectively blocked the effects of apigenin on induction of GSH depletion and apoptosis in MG-63 cells. Conclusion Apigenin induces apoptosis in human osteogenic sarcoma MG-63 cell line through GSH depletion.
