胎盘脂溶性小分子化学成分

摘    要:目的研究人胎盘的脂溶性小分子化学成分。方法采用硅胶柱色谱、SephadexLH-20凝胶柱色谱等方法进行分离纯化,通过波谱分析和单晶衍射法进行结构鉴定。结果从胎盘中分离鉴定了21个化合物,分别是环(亮-丙氨酸)(1),环(缬-亮氨酸)(2),环(脯-亮氨酸)(3),环(脯-缬氨酸)(4),环(亮-苯丙氨酸)(5),咖啡因(6),尿嘧啶(7),胸腺嘧啶(8),2’-甲氧尿苷(9),2’-脱氧尿苷(10),胆固醇(11),16α-羟基孕甾酮(12),16α-羟基睾酮(13),雌三醇(14),20(尺)-羟基黄体酮(15),烟酰胺(16),4-(2’-羟基苯基)-丁烯酮(17),N-棕榈酰鞘氨醇(18),N-十八烷基鞘氨醇(19),棕榈酸甘油单酯(20),甘油硬脂酸酯(21)。结论化合物1—5和17为首次从人体胎盘中分离得到,也是首次在人体中发现。

Abstract:Objective To investigate the lipophilic constituents of human placenta. Methods The compounds were separa- ted and purified by various column chromatography( silica gel,Sephadex LH-20) and their structures were established by spectroscopic methods and X-ray diffraction analysis. Rusults Twenty-one compounds were isolated from human placenta. Their structures were identified as cyclo (Leu-Ala) ( 1 ), eyclo (Leu-Val) (2), cyclo (Leu-Pro) ( 3 ), cyclo (Val-Pro) ( 4 ), cyclo (Phe-Leu) ( 5 ), caffeine ( 6 ), uracil( 7 ), thymine ( 8 ), 2'-O-methyluridine ( 9 ), 2'-deoxyuridine ( 10 ), cholesterol ( 11 ), 16α-hydroxyprogesterone ( 12 ), 16α- hydroxytestosterone ( 13), estriol ( 14 ), 20 (R) -hydroxypregn-4-en-3-one ( 15 ), nicotinamide ( 16 ), 4- ( 2'-hydroxyphenyl ) - (E) -3-buten- 2-one ( 17 ), N-palmitoyl-D-erythro-sphingosine ( 18 ), N-octadecanoyl-D-erythro-sphingosine ( 19 ), 1 -monopalmitin ( 20 ), and 1 -monoste- arin (21). Conclusion The cyclic dipeptides 1-5 and 17 were isolated from human placenta for the first time. They were also firstly i- dentified from human body.
