小檗碱对去卵巢大鼠骨量丢失的保护作用

摘    要:目的:研究小檗碱对去卵巢大鼠骨量丢失的作用.方法:以去卵巢诱导大鼠骨质疏松症,3月龄雌性SD大鼠随机分为假手术组、去卵巢组(空白组)、阳性对照组(尼尔雌醇组)及低(10mg· kg^-1)、中(30 mg· kg^-1)、高(90 mg· kg^-1)剂量小檗碱组,每组10只.12周后,取胫骨近端行不脱钙制片,采用骨组织形态计量学方法测定胫骨中段皮质骨动、静态学参数.双能X射线骨密度仪测量股骨骨密度.结果:与空白组比较,小檗碱能显著增加大鼠胫骨中段松质骨骨小梁面积百分比和骨小梁厚度(P<0.01),减少骨小梁分离度(P<0.05);增加骨形成参数:矿化沉积率(MAR)、骨形成率(BFR/BS、BFR/BV、BFR/TV)(P<0.05或0.01);同时,显著增加去卵巢大鼠股骨骨密度(P<0.05).对胫骨干骺端松质骨破骨细胞数目及体质量差异无统计学意义.结论:小檗碱对去卵巢大鼠骨量丢失有保护作用,主要是通过促进骨形成、抑制骨吸收、提高骨密度.

Abstract:Objective:To study the effect of berberine on bone loss in ovariectomized rats.Methods:The osteoporosis model was induced by ovariectomy.Three-month female Sprague-Dawley rats were randomly divided into six groups:sham-operation group,ovariectomized rats group(control-group),positive control group(nylestriol group),low-dose berberine (10 mg · kg^-1) group,mediumdose berberine (30 mg · kg^-1) group and high-dose berberine (90 mg · kg^-1) group.Each group had ten rats.After 12 weeks,the undecalcified longitudinal proximal tibial metaphysical sections were cut and stained for the bone histomorphometric static and dynamic parameters analysis.Bone density of femur metaphysis was determined by dual-energy X-ray absorptiometry.Results:Compared with the control group,berberine could significantly increase cancellous bone trabecular area and trabecular thickness in middle tibia (P 〈0.01) and decrease trabecular separation(P 〈 0.05).The parameters of bone formation such as MAR,BFR/BS,BFR/BV and BFR/TV were also obviously higher than those of control group(P 〈 0.05,P 〈 0.01).Meanwhile,berberine could enhance bone density of femur metaphysis in ovariectomizedrats significantly(P 〈 0.05).There was no significant change in osteoblasts number of tibial metaphyseal cancellous bone and body.Conclusion:Berberine shows prevention effect on bone loss in ovariectomized rats by promoting bone formation,increasing bone density and inhibiting bone reabsorption.
