摘    要:目的 建立测定罗勒多糖的分子量与分子量分布的方法.方法 应用高效凝胶色谱法,色谱柱为Shodex SB-804HQ凝胶色谱柱,检测器为示差折光检测器,流动相为0.7%硫酸钠溶液(含0.02%叠氮化钠),柱温为35℃,流速为0.5 mL/min.结果 6批样品的重均分子量分布在80 000-100 000之间,精密度、重现性良好.结论 试验方法快速、准确,结果可靠,为质量控制提供了基础.

Abstract:Objective To establish a method for determining the molecular weight (Mw) and weight distribution (Mw distribution) in basil polysaccharide by high performance gel chromatography(HPGC). Methods Using HPGC method,the chromatographic condition in- cluded the Shodex SB-804 HQ gel chromatographic column,the mobile phase was 0.7% Na2SO4 (containing 0. 02% NAN3) at the column temperature of 35℃ and the flow rate of 0.5 mL/min with the RID detector. Results Mw of the basil polysaccharide samples were 80 000- 100 000 with good precision and good reproducibility. Conclusion The established method is rapid,accurate and sensitive for testing Mw and Mw distribution in basil polysaccharide by HPGC.
