摘    要:目的建立同时测定十味活血丸中升麻苷和甘草苷含量的HPLC方法。方法采用HPLC法,色谱柱为D iamonsil C18(200 mm×4.6 mm,5μm),流动相为乙腈-体积分数为0.5%的冰醋酸溶液(体积比为15∶85),流速:1.0 mL.m in-1,检测波长:276 nm,柱温:40℃。结果十味活血丸中升麻苷和甘草苷与其他成分分离良好。升麻苷的质量浓度在2.86~28.6 mg.L-1(r=0.999 7)、甘草苷的质量浓度在0.774~7.74 mg.L-1(r=0.999 9)内与峰面积呈良好的线性关系,升麻苷和甘草苷的平均回收率分别为99.2%和99.0%,RSD(n=9)分别为1.8%和1.6%。结论该法可用于十味活血丸中升麻苷和甘草苷的含量测定。

Abstract:Objective To establish an HPLC method for simultaneous determination of prim-O-glucosylcimifugin and liquiritin in Shiweihuoxue(traditional Chinese medicine) pill. Methods The separation was performed on a Diamonsil C18 (4.6 mm × 200 mm, 5 μm)column. The mobile phase was composed of acetonitrile and 0.5 % (Ф) glacial acetic acid solution at the rate of 15:85. The column temperature was set at 40 °C and the flow rate was 1.0 mL &;#8226; min-1, with UV detection at 276 nm. Results The numbers of theoretical plates calculated for prim-O-glucosylcimifugin and liquiritin were 4600 and 5500, respectively. The regression equation of prim-O-glucosylcimifugin was A = 8.910 × 103ρ + 1.812 × 10(r = 0.999 7), which was linear over the ranger of 2.86-28.6 mg&;#8226;L-1. The regression equation of liquiritin was A = 2.548 × 104ρ + 8.741 × 10(r = 0.999 9), which was linear over the ranger of 0.774-7.74 mg&;#8226;L-1.The average recovery was 99.2 % (RSD = 1.8 % ) for prim-O-glucosylcimifugin, 99.0 % (RSD = 1.6 %) for liquiritin. Conclusion The method is simple and accurate, and can be used as the method for quality assessment of Shiweihuoxue pill.
