摘    要:目的建立小承气汤中大黄酸和厚朴酚的HPLC含量测定方法。方法样品以体积分数为70%的乙醇回流提取,采用Thermo C18柱(250 mm×4.6 mm,5μm),流动相为甲醇体积分数为1%乙酸水溶液(体积比为72∶28),流速为1.0 mL.min-1,检测波长为254 nm。结果大黄酸和厚朴酚的线性范围分别为1.52~19.0 mg.L-1和1.68~21.0 mg.L-1,平均回收率为97.6%和101.3%。结论建立的方法可用于小承气汤的质量控制。

Abstract:Objective To establish a reversed-phase HPLC method for determination of rhein and magnolol in Xiaochengqi decoction.(tradi tional Chinese medi cines)Methods The sample was separated on a Thermo-C_(18) column(250 mm×4.6 mm,(5 μm))using methanol-1% acetic acid solution(V∶V=72∶28) as the mobile phase at a flow rate of 1.0 mL·min~(-1).The detection was set at 254 nm.Results The calibration curve was linear in the range of 1.5219.0 mg·L~(-1)(r=0.999 7) for the rhein and 1.68-21.0 mg·L~(-1) for the magnolol(r=0.999 6).The inner-day RSD was 0.15% and 0.14%,and the average was 97.6% and 101.3% respectively.Conclusions This method is accurate,simple and sensitive,and can be used for the quality control of Xiaochengqi decoction.
