摘    要:目的建立同时测定十味益脾颗粒中绿原酸与芥子碱硫氰酸盐含量的方法。方法采用HPLC法。色谱柱为Kromasil 100A C18柱(200 mm×4.6 mm,5μm),流动相为乙腈-0.08 mol.L-1磷酸二氢铵梯度洗脱;流速为1.0 mL.min-1,柱温为30℃,检测波长为326 nm。结果绿原酸和芥子碱硫氰酸盐分别在0.858~21.500 mg.L-1(r=0.999 8)、2.140~53.600 mg.L-1(r=0.999 8)内与峰面积呈良好的线性关系;平均方法回收率分别为98.7%(RSD=1.7%,n=9)和99.0%(RSD=1.9%,n=9)。结论 HPLC法可用于同时测定十味益脾颗粒中绿原酸和芥子碱硫氰酸盐的含量。

Abstract:Objective To develop an HPLC method for the simultaneous determination of chlorogenic acid and sinapine thiocyanate in Shiweiyipi granula(traditional Chinese medicines).Methods Chromatography was performed on a Kromasil100A C18 column(200 mm×4.6 mm,5 μm)with the mobile phase of acetonitrile-0.08 mol · L-1 ammonium dihydrogen phosphate solution,gradient elution.The flow rate was 1.0 mL · min-1.The column temperature was 30 ℃ and the detection wavelength was set at 326 nm.Results The linear calibration curve were obtained in the concentration range of 0.858-21.500 mg · L-1for chlorogenic acid and 2.140-53.500 mg · L-1for sinapine thiocyanate.The mean recoveries of chlorogenic acid and sinapine thiocyanate were 98.7%(RSD=1.7%,n=9)and 99.0%(RSD=1.9%,n=9),respectively.Conclusions The method can be used to control the quality of Shiweiyipi granula,which provides the advantage of quickness,simplicity and repeatability.
