2-氨基磷酸茚和茉莉酸甲酯对高山红景天愈伤组织生长和红景天苷积累的影响

目的考察2-氨基磷酸茚(2-aminoindan-2-phosphate,AIP)和茉莉酸甲酯(methyl jasmonate,MJ)对高山红景天愈伤组织生长和红景天苷含量的影响。方法将培养10 d的高山红景天愈伤组织,分别接种在含有不同浓度的AIP和MJ的固体培养基中,每两天取样一次,测定愈伤组织生物量和红景天苷含量的变化趋势。结果 AIP和MJ最佳浓度分别为10μmol.L-1和200μmol.L-1时红景天苷含量达到最大值,分别为6.52 mg.g-1和5.98 mg.g-1,是空白对照组的1.43和1.32倍;愈伤组织中红景天苷的含量均高于野生药材中的含量。结论 AIP和MJ可显著提高红景天苷含量,高浓度的MJ和AIP对高山红景天愈伤组织的生长有抑制作用。

Effects of methyl jasmonate and 2-aminoindan-2-phosphate treatment on growth and salidroside accumulation in callus culture of Rhodiola sachalinensis A. Bor.

Objective To study the effects of methyl jasmonate(MJ)and 2-aminoindan-2-phosphate(AIP)on callus growth and salidroside accumulation of Rhodiola sachalinensis A.Bor.callus. Methods Callus on the 10th day culture was inoculated on subculture medium containing different initial concentration of MJ and AIP.They were sampled every two days for dry weight and salidroside content determination. Results The optimal concentrations of AIP and MJ were 10 and 200 μmol·L-1,respectively.Salidroside content reached 6.52 and 5.98 mg·g-1,respectively,which was 1.43-fold and 1.32-fold of that of control group,respectively. Conclusions Both AIP and MJ can significantly improve salidroside accumulation.Higher concentrations of MJ and AIP show negative effects on callus growth.
