阿卡波糖产生菌代谢调控的研究

摘    要:目的探索4种可能的调节物对阿卡波糖生物合成的调节作用,以提高阿卡波糖的发酵产量。方法根据阿卡波糖的生物合成途径,在阿卡波糖发酵的不同阶段添加不同浓度的代谢调节物质,依据阿卡波糖的发酵水平确定添加代谢调节物质的最佳时间和最佳质量浓度。结果 0 h于发酵培养基中添加1 mg.L-1碘乙酸,发酵结束时阿卡波糖的发酵单位提高了8.9%;48 h添加1 mg.L-1NaF,发酵单位提高了13.3%;72 h添加0.5 g.L-1柠檬酸钠,发酵单位提高了6.2%;48 h添加2 g.L-1莽草酸,发酵单位提高了17.3%。结论这4种化合物对阿卡波糖的生物合成都有一定的调节作用,在最佳的时间以最适的浓度添加可以提高阿卡波糖的发酵产量。

Abstract:Objective To investigate the effects of four possible regulators on the biosynthesis of acarbose in order to increase the fermentation yield of acarbose. Method According to the biosynthesis pathway of acarbose, four kinds of possible metabolism regulators were fed at different stages of the acarbose fermentation on different concentrations; the optimal feeding time and concentration were determined according to the final fermentation yield of acarbose. Results Results showed that the productivity of acarbose was increased by 8.9%, when 1 mg·L-1 iodoacetic acid was added into the medium at 0 h; the yield was increased by 13.3%, when 1 mg·L-1 sodium fluoride was added at 48 h; the productivity was increased by 6.2%, when 0.5 g·L-1 sodium citrate was added at 72 h; the yield was increased by 17.3%, when 2 g·L-1 shikimic acid was added at 48 h. Conclusions The four kinds of chemicals have regulatory effects on the biosynthesis of acarbose; the fermentation yield of acarbose can be increased if the chemicals are fed at a suitable time and optimal concentration.
