双参通脉滴丸对缺血再灌注损伤性大鼠脑梗死面积及神经细胞凋亡的影响

摘    要:目的:研究双参通脉滴丸对大鼠脑缺血再灌注损伤脑组织梗死面积和神经细胞凋亡的影响及该影响是否存在剂量依赖性。方法:采用双侧颈总动脉阻断的方法建造大鼠脑缺血再灌注损伤模型。将大鼠随机分为假手术组、模型组、双参通脉滴丸(高、中、低剂量)组。采用TTC染色法测定脑组织梗死面积,末端脱氧核苷酸转移酶介导的dUTP缺口末端标记法(TUNEL)检测脑组织凋亡神经细胞。结果:双参通脉滴丸(高、中、低剂量)组TTC染色法测定脑组织梗死面积与模型组比较差异显著,与假手术组无统计学意义;采用TUNEL法检测脑组织凋亡神经细胞数与模型组比较有差异,与假手术组相近。结论:双参通脉滴丸能减轻大鼠脑缺血再灌注损伤后脑组织梗死面积,抑制脑神经细胞的凋亡,并且其作用存在剂量依赖性。

Abstract:Objective:To study the effects of Shuangshen Tongmai dripping pills(STD)on cerebral infarction size and neuronal apoptosis after cerebral ischemiare-perfusion injury(IRI)in rats and to know whether or not it shows a dose-dependent relationship.Methods:The bilateral carotid artery occlusion method was used to establish rats cerebral ischemia-reperfusion injury mode1.All of rats were divided into five groups randomly:sham group,model group,STD high-dose treatment group,STD medium-dose treatment group,and STD low-dose treatment group.The TTC staining was used to measure brain infarction area,and the terminal deoxvnudeotidv1 transferase meditated Dutp-nickend labeling(TUNEL)was used to determine brain nerve cells apoptosis.Results:The brain infarction size of SSTMDP high,medium,low dose treatment group had significant differences from the model group,and were not significant from the sham group.The apoptosis brain nerve cells of SSTMDP high,medium,low dose treatment group had significant differences from model group,the sham group.Conclusion:STD can reduce the rats cerebral ischemia-reperfusion injury cerebral infarction area,and inhibit brain cell apoptosis.It shows a dose-dependent relationship.
