晚期转移性乳腺癌血浆MMP-2、MMP-9检测与化疗疗效相关性研究

摘    要:目的 探讨血浆基质金属蛋白酶活性与晚期转移性乳腺癌化疗疗效的相关性。方法 选择经病理或细胞学确诊的晚期转移性乳腺癌患者,分别于化疗前后收集患者血浆(1.8mL空腹全血加0.2%mL枸橼酸钠),离心后放置于-70℃冰箱冻存备查。用酶谱法测定肿瘤患者血浆MMP-2及MMP-9的活性,用凝胶图象处理系统分析;以白色条带的净光密度值相对定量样品中酶的活性。结果 MMP-2、MMP-9在晚期乳腺癌治疗前后的活性水平测定具有统计学意义。(P<0.05)。结论 晚期乳腺癌患者血浆MMP-2、MMP-9活性与病情进展和疗效有一定相关性,而检测方法简单经济,具有一定的临床指导价值。

Abstract:Objective To evaluate the bioactivity of plasmatic MMPsand their relevance with chemotherapeutic efficacy in patients with advanced metastatic breast cancer.Methods Plasma samples of patients with pathological or cytological confirmed advanced metastatic breast cancer were collected before and after chemotherapy.The Zymography method was used to determine the bioactivity of MMP-2 and MMP-9,followed by systemic analysis with gel imaging procedure.The optic density of white band was taken as the corresponding bioactivity of quantitative samples.Results The level of MMP-2,MMP-9 in the activity of advanced breast cancer before and after treatment were statistically significant(P < 0.05).Conclusion The bioactivity of MMP-2 and MMP-9 in plasma in patients with MBC before and after chemotherapy has a certain relationship with disease progression and chemotherapeutic efficacy.The detective method is economic and have clinical guidance value.
