高效液相色谱法测定参芪十一味颗粒中橙黄决明素和大黄酚的含量

目的建立测定参芪十一味颗粒中橙黄决明素和大黄酚的高效液相色谱方法。方法选用Agela venusil MP C18色谱柱（250mm×4.6mm,5μm）,流动相：A相为乙腈,B相为0.05%磷酸溶液;梯度洗脱：0~15min,60%B~60%B,15~30min,60%B~90%B,30~40min,90%B~90%B;流速：1m L·min^-1,检测波长284nm。结果样品中两种成分得到良好分离,橙黄决明素在0.00245~0.0735mg·m L^-1（r=0.9999）、大黄酚在0.00284~0.0851mg·m L^-1（r=0.9999）范围内线性关系良好,回收率分别为100.51%、101.38%,RSD分别为1.11%、1.99%（n=6）。结论该方法简单准确,专属性强,重复性好,可用于参芪十一味颗粒的质量控制。

Determination of Aurantio-obtusin and Chrysophanol in Shengqi Shiyiwei Granules by HPLC

OBJECTIVE To establish a HPLC method for the determination of Aurantio-obtusin and Chrysophanol in Shengqi Shiyiwei Granules. METHODS The determination was performed on an Agela venusil MP C18 column. The mobile phase consisted of（A） ACN（B） 0. 05% H3PO4 solution with the gradient elution 0 ~ 15 min,60% B ~ 60% B,15 ~ 30 min,60% B ~ 90% B,30 ~ 40 min,90% B ~ 90% B. The flow rate was 1m L·min^-1,and the detection wavelength was set at 284 nm. RESULTS The method showed a good linear relationship within the range of 0. 00245 ~ 0. 0735mg·m L^-1for Aurantio-obtusin（r = 0. 9999）,0. 00284 ~ 0. 0851mg·m L^-1for Chrysophanol（r = 0. 9999）,The average recovery were 100. 51%（RSD was 1. 11%） for Aurantio-obtusin and 101. 38%（RSD was 1. 99%） for Chrysophanol（n = 6）. CONCLUSION The method is simple,accurate,sensitive and can be used for the determination of Aurantio-obtusin and Chrysophanol in Shengqi Shiyiwei Granules.
