摘    要:目的:优化白及多糖的提取工艺。方法:加热回流法提取白及多糖,应用UV测定含量并计算白及多糖得率。以正交试验其中的7组数据作为训练数据样本,剩余2组数据作为验证数据样本,采用误差反向传播人工神经网络(BP-ANN)进行建模及分析,以白及多糖得率作为考察指标,仿真模拟预测白及多糖最佳提取工艺参数。结果:正交试验优化的提取工艺为加12倍量水煎煮3次,每次煎煮2 h,白及多糖得率为24.70%;BP-ANN预测的最佳提取工艺为加16倍量水煎煮3次,每次煎煮1.5 h,白及多糖得率为29.31%。验证试验结果表明,BP-ANN优化工艺与正交试验相比,白及多糖得率的增长率为16.07%,与预测值的相对误差为2.18%。结论:BP-ANN优化工艺提取效率高于正交试验,BP-ANN仿真预测的提取工艺稳定、白及多糖得率高。

Abstract:OBJECTIVE To optimize the extraction process of polysaccharides from Bletillae Rhizoma.METHODS The polysaccharides were extracted from Bletillae Rhizoma by heating reflux method and determined by UV,and the yield of polysaccharides from Bletillae Rhizoma was calculated. The datas of seven groups in the orthogonal test were used as the training data samples, and those in other two groups were used as the verification data samples, BP-ANN (error back propagation artificial neural network) was used for modeling and analysis, and the yield of polysaccharides from Bletillae Rhizoma was used as the index to simulate and predict the optimal extraction process parameters of polysaccharides from Bletillae Rhizoma.RESULTS The orthogonal test was designed to optimize the extraction technology: decocting 3 times with 12-fold amount of water, decocting 2 h for each time, and obtaining 24.70% of polysaccharides from Bletillae Rhizoma; BP-ANN predicted that the best extraction process was to add 16 times of water for 3 times, and 1.5 h for each time, and the yield of polysaccharides from Bletillae Rhizoma was 29.31%. The results of validation test showed that the optimized process of BP-ANN was better than that of orthogonal test, and the increasing rate of the yield of polysaccharides from Bletillae Rhizoma was 16.07% with the relative error of 2.18%.CONCLUSION The extraction efficiency of BP-ANN optimization process is higher than that of orthogonal test, BP-ANN simulation predicted that the extraction process was stable and the yield of polysaccharides from Bletillae Rhizoma was high.
