理气散结汤对大鼠乳腺增生的治疗作用及其机制

目的：研究理气散结汤对大鼠乳腺增生的治疗作用，并对其作用机制进行初步探讨。方法：采用肌内注射雌-孕激素联合诱导制备乳腺增生模型大鼠，选择造模成功的大鼠按体质量随机分为模型组、小金胶囊组、理气散结汤低、中、高剂量组，另设正常对照组。连续给药30 d后，测定各组大鼠胸腺、脾脏和子宫重量系数，观察乳腺组织病理形态改变，ELISA法测定血清雌二醇（E₂）和黄体酮（P）含量，RT-PCR检测雌激素受体α（ERα）基因表达水平，Western-blot检测ERα蛋白表达水平。结果：与模型组比较，理气散结汤能显著增加乳腺增生大鼠脾脏和胸腺重量指数（P<0.05），减小子宫重量指数（P<0.05），显著减轻大鼠乳腺增生程度，显著降低血清E₂含量（P<0.05）、增加P的含量（P<0.05），显著下调ERα基因和单边表达水平（P<0.05）。结论：理气散结汤对大鼠乳腺增生有良好的治疗作用，其机制与调节乳腺增生大鼠血清中性激素及其受体水平有关。

Therapeutic effects and underly mechanism of Liqi Sanjie decoction against hyperplasia of mammary glands in rats

OBJECTIVE To study the therapeutic effects of Liqi Sanjie decoction against hyperplasia of mammary glands in rats, and explore the underly mechanism. METHODS The model of hyperplasia of mammary gland was induced by estrogen and progestogen injected intramuscularly. Rats successfully modeled were randomly divided into model group, Xiaojin capsule group, Liqi Sanjie decoction high, medium and low dose groups. Normal control group was also alloated. After 30 days of continuous administration, thymus, spleen and uterine weight coefficients of rats were observed, pathological changes of breast tissues were detcted, estradiol (E₂) and progesterone (P) contents were detected via ELISA, the expression of hormone receptor α (ERα) gene was detected by RT-PCR, and the protein expression of ERα was detected by Western-blot.RESULTS Compared with model group, Liqi Sanjie decoction significantly increased spleen and thymus mass index in rats with breast hyperplasia (P<0.05), reduced uterine mass index (P<0.05), significantly reduced the degree of rat mammary gland hyperplasia and E₂ content (P<0.05), increased P content (P<0.05), significantly reduced gene and protein expression of ERα (P<0.05).CONCLUSION Liqi Sanjie decoction has good therapeutic effects against hyperplasia of rats mammary glands, and the underly mechanism may be related with regulating levels of sex hormones and its receptors.
