摘    要:建立了何首乌中的主要有效成分大黄素、大黄素甲醚、大黄酚及大黄酸等蒽醌类的反相高效液相色谱法。以TSD-gel80Ts为固定相,甲醇-水-磷酸(720:280:1)为流动相。样品用70%甲醇提取浓缩后用氯仿萃取,分离出游离型和结合型蒽醌的定量用试液。本法精确、可靠。用此方法测定了不同产地的9种何首乌和4种夜交藤样品。

Abstract:High performance liquid chromatographic method was developed for the determination of free and glycosidic anthraquinones, namely rhein, emodin, chrysophanol and physcion and their glycosides, in Polygonum multiflorum. A reversed-phase system consisting of a chemically bonded ODS silica gel column with a mixture of methanol-water-phosphoric acid (720 : 280 : 1) as the mobile phase was used. The sample was extracted by supersonic with 70%methanol and concentrated, then partitioned with chloroform and water to separate free and glycosidic anthraquinone fractions. Furthermore, glycosidic anthraquinone fraction was hydrolysed to free form. Market samples were determined by this method. Emodin, physcion and their glycosides were found in Polygonum multiflorum, but the other anthraquinones were not detected.
