摘    要:目的建立中药黄芪以及黄芪发酵液中有机硒和无机硒的含量测定方法。方法采用微波消解和三羟甲基氨基甲烷-盐酸(Tris-HCl)缓冲溶液提取后获得总硒和无机硒,并用原子荧光光谱法测定总硒和无机硒的含量,差值即为有机硒的含量。结果硒的质量浓度在0~100μg·L~(-1)范围内与测定值呈线性变化,检出限为0.090 4μg·L~(-1),回收率为94%~104%。结论该方法简单、灵敏、快速,可作为中药及其发酵液或萃取液中硒含量的测定方法。

Abstract:Objective This research aimed to determinate the organic selenium and inorganic selenium in Radix Astragali and its fer-mented liquid.Methods Microwave digestion and HCl solution with Tris solution(Tris-HCl)extraction were used to obtain the to-tal selenium and inorganic selenium,and atomic fluorescence spectrometry was used for determining the content of total selenium and inorganic selenium,the difference was the organic selenium.Results The linear range of the method was 0-100μg·L-1 and the limit of detection was 0.0904μg·L-1 .The range of recovery rate was 94%-104%.Conclusion This method is simple and quick,and can be used to determinate the trace selenium in the herbal drugs.
