摘    要:目的:建立同时测定益气祛白颗粒中黄芪甲苷Ⅳ和木通皂苷D含量的方法。方法:采用高效液相色谱法。色谱柱为YMC-Pack ODS-AM(150 mm×4.6 mm,3μm),流动相为乙腈-水(梯度洗脱),流速为0.8 ml/min,检测波长为203 nm。结果:黄芪甲苷Ⅳ和木通皂苷D的质量浓度分别在10500、2.5500、2.5250μg/ml范围内与各自峰面积积分值呈良好的线性关系(r分别为0.999 9和0.999 7);平均加样回收率分别为106.09%、99.45%,RSD分别为1.81%、3.24%(n均为6)。结论:所建方法简单、稳定,结果准确、可靠,可为益气祛白颗粒的体内药动学研究提供依据。

Abstract:OBJECTIVE: To develop a method for simultaneous determination of astragaloside Ⅳ and akebia saponin D in Yiqi Qubai granules. METHODS: HPLC method was adopted. The determination was performed on a YMC-Pack ODS-AM (150 min×4.6mm, 3μm) column with mobile phase consisted of acetonitrile-water (gradient elution) at the flow rate of 0.8 ml/min. The detection wavelength was set at 203 nm. RESULTS: The linear ranges were 10-500 μg/ml for astragaloside Ⅳ (r:0.999 9) and 2.5-250 μg/ml for akebia saponin D (r= 0.999 7). The average recoveries were 106.09 % (RSD: 1.81%, n= 6) and 99.45 % (RSD= 3.24%, n=6), respectively. CONCLUSIONS: The established method is convenient, stable, accurate and reliable, and can provide reference for pharmacokinetic study of Yiqi qubai granules in vivo.
