乳胶过敏原Hev b1二级结构及抗原表位的预测

摘    要:目的利用生物信息学软件预测乳过敏原Hevb1的二级结构及B细胞和T细胞的抗原表位。方法在Uniprot数据库中获得Heyb1蛋白序列,通过GOR4在线软件预测其二级结构;采用DNAStar的Protean模块预测Hevb1蛋白的亲水性、表面可及性、柔韧性及抗原指数,从而综合预测其B细胞表位;应用NetMHCⅡ2.2在线服务器预测Hevb1蛋白的T细胞表位。结果乳胶过敏原Heyb1的B细胞优势抗原表位为1—14,40一46;T细胞优势抗原表位为56—64,76—84,97—116。结论通过生物信息学分析获得乳胶过敏原Hevb1的B细胞和T细胞优势抗原表位,可为乳胶过敏原Hevb1的抗体设计、过敏原性改浩等研奔打下略宴的基础.

Abstract:Objective To predict the secondary structure and B/T cell epitopes of Hey b 1 protein for latex by application of bioinformatics software. Methods The amino acid sequence of Hev b 1 was obtained from the Uniprot database. The secondary structure of Hey b 1 was predicted by the GOR 4 online server. DNAStar Protean was applied to predict the B cell epitopes by analyzing the hydrophilicity, surface probability, flexible regions and antigenic index of Hev b 1, and the T cell epitopes were predicted through the NetMHC Ⅱ 2.2 server. Results The dominant B cell epitopes of Hev b 1 were on the regions of 1-14 and 40-46 and the dominant T cell epitopes were on 56-64, 76- 84 and 97-116. Conclusions We obtain the dominant B/T cell epitopes of latex allergen Hev b 1 by bioinformatic analysis, which lays a solid foundation for designing antibodies and modifying allergenicity of Hev b 1.
