普罗布考与阿托伐他汀联合用药对高脂饮食自发性高血压大鼠炎症因子的影响

摘    要:目的:对普罗布考和阿托伐他汀联合用药在高脂饮食自发性高血压大鼠(SHR)的抗炎作用进行评价。方法:92只雄性SHR随机分为5组,C组:普通饲料+生理盐水;M组:高脂饲料+生理盐水;P组:高脂饲料+普罗布考(150mg·kg^-1·d^-1);A组:高脂饲料+阿托伐他汀(10mg·kg^-1·d^-1);P+A组:高脂饲料+阿托伐他汀(10mg·kg^-1·d^-1)+普罗布考(150mg·kg^-1·d^-1)。实验8周后处死SHR抽查血脂(TC、TG、LDL-C)和血清炎症指标(IL-6、TNF-α、hs—CRP)并与实验前各指标作比较。结果:高脂饮食各组血清TC、LDL—C水平较基础值明显升高且以M组最高(均P〈0.01),TG下降(P〈0.01)。各用药组(尤其P+A组)TC、LDL—C明显低于M组(均P〈0.01),血清TG水平P+A组与A组均低于M组(均P〈0.01);血清IL-6、TNF—α、hs—CRP水平变化中,M组较基础值、C组明显升高(均P〈0.01),P组、A组、P+A组较M组明显降低(均P〈0.01)。P+A组较单独用药两组降低更明显(P〈0.01).结论:高脂饮食可导致血脂、炎症因子的紊乱;普罗布考与阿托伐他汀联合用药在增强其调脂作用的同时,也增强其抗炎作用,且效果优于二者单独用药.

Effects of Combination Therapy of Probucol and Atorvastatin on Adjusting Inflammatory Factors in Spontaneously Hypertensive Rat with High Lipids Diet

Abstract:Objective: To evaluate the effects of combination therapy of probucol and atorvastatin on adjusting inflammatory factors in spontaneously hypertensive rat (SHR) with high lipids diet. Methods: 92 SHRs were randomly divided into 5 groups. Group C was treated with normal diet and normal saline. Group M, Group A, Group P and Group P + A were all treated with high lipids diet. At the same time, group M, group P, group A, group P +A were administrated with normal saline, probucol (150 mg·kg^-1·d^-1)), atorvastatin (10mg·kg^-1·d^-1)), and probucol (150 mg·kg^-1·d^-1)) plus atorvastatin (10 mg·kg^-1·d^-1)), respectively. After 8 weeks treatment, all rats were killed. The serum levels ofTC, TG, LDL-C, IL-6, TNF-α and hs-CRP were measured respectively before and after the experiment. All the above indexes were compared intra-groups and inter-groups. Results: Serum levels of TC, LDL-C in high lipids diet group (especially in group M) were obviously higher than baseline data (P 〈 0.01), but serum levels of TG decreased. Serum levels of TC and LDL-C in group P, group A and group P + A were dramatically lower than that of group M (P 〈 0.01), especially in group P + A. Serum levels of TG in group P + A and group A were lower than that of group M (P 〈 0.01). Serum levels of IL-6, TNF-α and hs-CRP in group M obviously higher than that of group C and baseline data (P 〈 0.01), and the inflammatory factors in group P, group A, especially group P + A were obviously lower than that of group M (P 〈 0.01). Conclusion: High lipids diet may lead to the disorder of serum lipids and inflammatory factors. Combination therapy of probucol and atorvastatin can strengthen the effects on adjusting lipids and decrease the levels of inflammatory factors, and the effects of combination therapy are stronger than that of monotherapy.
