摘    要:目的:建立测定蜘蛛香药材中4种缬草素类成分缬草三酯、Chlorovaltrate、Jatamanvaltrate P和Jatamanvaltrate Q含量的HPLC法。方法:采用Eclipse OB-C18色谱柱(250 mm×4.6 mm,5 μm);以乙腈(A)-水(B)为流动相进行梯度洗脱,0~20 min A∶B=75∶25,20~25 min A∶B=87∶13,25 min后A∶B=75∶25;流速0.8 ml/min;检测波长256 nm;柱温25 ℃。结果:缬草三酯、Chlorovaltrate、Jatamanvaltrate P和Jatamanvaltrate Q的线性范围分别为0.18~3.06 mg、0.23~3.41 mg、0.12~1.72 mg、1.22~20.41 mg,精密度RSD分别为1.31%、1.41%、2.82%和1.35%(n=7),重现性RSD分别为2.08%、1.43%、1.86%和1.39%(n=4),平均加样回收率分别为(99.72±0.33)%、(97.74±1.05)%、(99.90±0.77)%和(100.03±1.47)% (n=5),稳定性RSD分别为2.78%、1.98%、2.21%和2.62%(n=3)。结论:本方法准确、简便、快速,可用于蜘蛛香药材的质量控制。

Simultaneous content determination of 4 valepotriate compounds in Valeriana jatamansi Jones by HPLC method

Abstract:Objective:To establish a HPLC method for simultaneous content determination of four valepotriate compounds (vahrate, Chlorovaltrate, Jatamanvaltrate P and Jatamanvaltrate Q) in Valeriana jatamansi Jones. Methods: The HPLC analysis was performed on an Eclipse OB-C18 column(250 mm× 4. 6 mm, 5 μm). The mobile phase was composed of acetonitrile(A)-water(B) in gradient elution at a flow rate of 0.8 ml/min,0-20 min A : B=75 : 25,20-25 min A : B=87 : 13,after 25 min,A : B=75 : 25. The detection wavelength was 256 nm and the column temperature was 25 ℃. Results:Vahrate,Chlorovahrate, J atamanvahrate P and J ata- manvaltrate Q were in good linearity within the ranges of 0.18-3.06 mg,0. 23-3. 41 mg,0. 12-1.72 mg and 1.22-20.41 mg,respective- ly. Precision RSD of the four compounds were 1.31%, 1.41%, 2.82% and 1.35% (n= 7) respectively; reproducibility RSD were 2.08% ,1.43% ,1.86% and 1.39%(n=4) ;average recovery rates were (99.72±0.33) %, (97.74±1.05)%, (99.90±0. 77) % and (100. 03±1.47)% (n=5) and the stability RSD were 2.78% ,1.98% ,2.21% and 2. 62%(n=3) respectively. Conclusion:The HPLC method is accurate, simple, rapid,and could be used for the quality control of Valeriana jatamansi Jones.
