益气通络法对糖尿病周围神经病变大鼠坐骨神经细胞凋亡和HO-1表达的影响

摘    要:目的]观察益气通络法对糖尿病大鼠周围神经病变(DPN)的疗效,并探讨其作用机制。方法]Wistar大鼠腹腔注射链脲佐菌素复制糖尿病模型,对成模大鼠每日用益气通络法灌胃,8周后检测坐骨神经细胞凋亡数目,乳酸脱氢酶(LDH)活性,促凋亡因子Bax、Caspase-3和血红素氧合酶-1(HO-1)的表达水平。结果]与模型组比较,益气通络法明显减少糖尿病大鼠坐骨神经凋亡细胞数目,降低LDH含量、抑制Bax、Caspase-3的表达,增加HO-1蛋白的表达。结论]益气通络法能有效上调HO-1的表达,降低坐骨神经细胞的凋亡,预防和治疗糖尿病周围神经损伤。

Abstract:Objective] To discuss the effects of Yiqi Tongluo formula on rats with diabetic peripheral neuropathy(DPN) and explore its potential mechanisms.Methods] The rat model of DPN was induced by intraperitoneal injection of streptozotocinith, and DPN rats were divided into treatment group treated with daily Yiqi Tongluo formula for eight weeks. The expression of LDH, Bax, Caspase-3 and the number of cell apoptosis in the sciatic nerve were evaluated and compared.Results] Compared with the model group, the number of apoptotic cells and the expression of LDH, Bax, Caspase-3 were lower in the Yiqi Tongluo group in rat sciatic nerve, while the expression of HO-1 is higher significantly.Conclusion] Yiqi Tongluo formula can effectively increase the expression of HO-1, inhibit the oxidative damage and reduce cell apoptosis in sciatic nerve.
