摘    要:目的:探讨西红花酸对心肌细胞氧化应激性损伤的效应。方法:采用过氧化氢(H2O2)诱导法制备大鼠培养心肌细胞氧化应激损伤模型,通过观察培养心肌细胞的形态,检测细胞培养液中乳酸脱氢酶(LDH)活性、细胞内丙二醛(MDA)含量和超氧化物歧化酶(SOD)活性及培养细胞台盼蓝摄取率,反映西红花酸对该模型的影响。结果:在本实验使用浓度范围内,H2O2损伤培养心肌细胞的作用呈浓度依赖性地增大;西红花酸则能减少该氧化应激细胞的形态学改变、降低培养液中LDH活性和胞内MDA含量、增加胞内SOD活性及减少细胞死亡数量。结论:西红花酸对H2O2,造成的培养心肌细胞氧化应激性损伤具有明显的保护作用。

Abstract:AIM: To observe the effect of crocetin on the damages induced by oxidative stress in myocardial cells. METHOD: Oxidative damage cell model was induced by H_2O_2. The morphological changes of the cells were observed, and LDH activities in the medium, intracellular MDA levels and SOD activities as well as the trypan blue absorbance rates of the cells were measured in the crocetin pretreated and unpretreated groups. RESULT: All the indexes tested changed to reflect the damage on the cells by H_2O_2, in a concentration-dependent manner among the doses used, but not so greatly in the crocetin pretreated groups compared with that in the unpretreated control. CONCLUSION: Crocetin can obviously protect cultured myocardial cells from oxidative damages induced by H_2O_2.
