九里香糖蛋白成分终止孕兔妊娠及其机理

摘    要:从九里香Murraya paniculata(L.)Jacks.茎皮中分离得到的糖蛋白成分给孕期12-16d的孕免ip10mg/kg或羊膜腔内注射3mg/胚胎,3-5d后获得明显的终止妊娠效果。同时给黄体酮1mg/kg连续6d不能对抗其抗孕作用。组织学检查蜕膜组织有变性、坏死,炎细胞浸润,血窦瘀血和出血等情况。本品对已孕和未孕小鼠离体子宫收缩无明显改变。在麻醉状态下,给孕兔iv10mg/kg时,5-6min后子宫呈张力增强性节律收缩。幼兔连续4d ip 20mg/kg不能对抗雌二醇和黄体酮所致的内膜增生反应。治疗量10mg/kg卵巢内妊娠黄体未见有特殊变化,指示终止妊娠不是抗黄体作用的结果。该糖蛋白既无雌激素样作用,又无抗HCG活性。在终止妊娠中可能是通过对蜕膜损害和导致PG释放而起作用。

Abstract:The effect of glycoprotein isolated from Murraya paniculata(L.) on days 12-16 of pregnant rabbits was shown to terminate pregnancy. When the glycoprotein was given to these experimental animals for 10 mg/kg by ip or 3 mg/embryon by intra-amniotic injection, the obvious abortion effects were obtained 3-5 days after treatment. The antipregnancy effect could not be antagonized by simultaneous administration of progesterone succesively for 6 days. By histological observation, the degeneration, necrosis, infiltration of leucocytes, blood stasis of sinusoid and homorrhage were found in decidual tissue. The glycoprotein did not influence the contraction of isolated non-pregnant and pregnant uterus in mice significantly. While the component is given iv to pregnant rabbits under anesthesia at a dose of 10 mg/kg, increased rhythmical contraction of uterine myometricum was observed. The glycoprotein of 20 mg/kg given to juvenile rabbits for 4 days could not inhibit the endometrial hyperplasia response induced by estradiol and progesterone. At the therapeutic dose of 10 mg/kg no significant changes were observed on the corpus lateum of gestation. It was indicated that antipregnancy effect of glycoprotein was not related to progesterone and glycoprotein had neither estrogen-like activity nor anti-HCG action. Our results suggest that glycoprotein probably produces antipregnancy effect by destruction of decidual tissue and release of PG.
