摘    要:目的:制备pH-时滞型布地奈德微丸并对其释药特性进行研究。方法:采用锅包衣法制备裁药微丸,并进行乙基纤维素有机溶液包衣及Eudragit FS30D水分散体包衣。用转篮法进行释放度实验,采用多种pH模式筛选和评价处方。结果与结论:包衣厚度及材料比和pH对释药速率均有影响。优选处方微丸在高低2种pH模式下,其释药特性符合设计要求。

Abstract:AIM: The aim of the present study was to develop a pH-dependent and controled-release micropellets of budesonide and to evaluate their in vitro release. METHODS: The drug loaded pellets were prepared and then coated with organic solution of EC and an new enteric polymer(Eudragit FS 30D). Dissolution tests were carried out in media simulating pH conditions at various locations in the gastro-intestinal tract. RESULTS: Dissolution rate was affected by coat thickness, ratio of the two polymers and the pH of medium. Experiments mimicking the extremes of GI conditions indicated that about 70% of the drug would be released to the ileum and proximal colon region. CONCLUSION: The pellets showed an in vitro dissolution profile more appropriate to the pH profile of the ileum and colon.It is promising for the therapy of Crohn's desease .
