甲钴胺、格列齐特及其联合用药对糖尿病大鼠周围神经病变的治疗作用

摘    要:目的:研究甲钴胺与格列齐特及其联合用药对链脲佐菌素(STZ)糖尿病大鼠周围神经病变的影响。方法:动物灌胃给药,连续8周,观察药物对链脲佐菌素糖尿病大鼠刺激痛闽、坐骨神经传导速度、糖化血红蛋白和糖化血清蛋白含量、血液流变学参数及坐骨神经醛糖还原酶活性和Na^ ,K^ —ATP酶活性等指标的影响。结果:与糖尿病模型组大鼠相比,各给药组大鼠尾机械刺激痛阈均显著性改善,坐骨神经传导速度明显性提高,醛糖还原酶活性降低,Na^ ,K^ —ATP酶活性提高;格列齐特及格列齐特与甲钴胺联合用药均可降低血液中的糖化蛋白水平;此外,格列齐特还可降低红细胞最大聚集指数;甲钴胺和格列齐特联合用药与两药单用相比,未观察到明显性差异。结论:甲钴胺与格列齐特及其联合用药对治疗STZ糖尿病大鼠周围神经病变具有治疗作用,但两药合用未显示出明显的疗效增强作用。

Abstract:To investigate the effects of methyl cobalamin, gliclazide and combined application of both on diabetic peripheral ne uropathy in streptozotocin diabetic rats. Drugs were administrated orally for 8 w eeks. Effects of drugs on pain threshold to mechanical stimulation, sciatic never conduction velo city, glycated hemoglobin and glycated serum protein content, rheologic paramete rs, aldose reductase activity and Na + ,K + ATPase activity of strepto zotocin d iabetic rats were determined. Compared with diabetic grou p, pain thresho ld to stimulation was improved, sciatic nerve conduction velocity was incre ased, aldose reductase activity was depressed and Na + ,K + ATPase activity was incr eased in each administrated group. Glycated hemoglobin and glycated serum protei n contents were decreased in gliclazide and combination group. In addition, eryt hrocyte maximal aggregation index was decreased in gliclazide group. Compared wi th methylcobalamin or gliclazide group, remarkable difference was not observed i n combination group. Methylcobalamin, gliclazide an d combined applic ation of both could prevent peripheral neuropathy in streptozotocin diabetic rats. More effective action was not observed in drug combination group.
