红景天甙防治兔肝移植术后疲劳综合征的实验研究

摘    要:目的 研究红景天甙降低兔肝移植术后疲劳综合征的机制.方法 日本大耳白兔30只,采用随机数表法分为假手术组、模型组、红景天甙组,每组各10只,采用小肠切除-吻合术模拟术后疲劳综合征动物模型,造模10 d后进行1/2减体积原位肝脏移植手术,术后红景天甙组经尾静脉注射红景天甙治疗7d.比较三组兔外周血传出神经递质5-羟色胺(5-HT)、垂体生长素(GH),大脑皮层电位及肝功能谷草转氨酶(AST)、胆红素(TBIL)、谷丙转氨酶(ALT)水平.结果 模型组造模10 d及治疗7 d时5-HT(9.93±0.22)ng/mL vs(16.23±0.52)ng/mL]、AST(504.7±37.9)U/L vs(554.3±32.9)U/L]、TBIL(54.87±3.21)μmol/L vs(76.87±4.22)μmol/L]、ALT(181.6±18.3)U/L vs(312.3±34.5)U/L]明显升高,GH(0.71±0.03)pg/mL vs(0.53±0.05)pg/mL]明显降低,大脑皮层放电(459.23±48.08)mV vs(513.03±50.28)mV]逐渐增强,与本组造模前及假手术组比较,差异均有统计学意义(P<0.05).红景天甙组造模10 d时5-HT(10.72±0.31)ng/mL]、AST(484.7±20.1)U/L]、TBIL(55.21±5.21)μmol/L]、ALT(186.2±19.2)U/L]明显升高,GH(0.75±0.02)pg/mL]明显降低,大脑皮层放电(451.71±45.58)mV]逐渐增强,与假手术组比较,差异均有统计学意义(P<0.05).治疗7 d时,红景天甙组兔5-HT(10.21±0.31)ng/mL]、大脑皮层电位(321.12±38.24)mV]、肝功能AST(194.3±22.1)U/L]、TBIL(24.07±1.25)μmol/L]、ALT(112.2±23.1)U/L]明显降低,GH(2.67±0.11)pg/mL]明显增高,与造模10 d及模型组比较,差异有统计学意义(P<0.05).结论 红景天甙可降低兔肝脏移植术后疲劳综合征.

Effect of salidroside on the prevention and treatment of fatigue syndrome after liver transplantation in rabbits

Abstract:Objective To study the mechanism of salidroside in reducing fatigue syndrome after liver trans-plantation in rabbits. Methods A total of 30 rabbits were randomly divided into the sham group, model group and sali-droside group according to random number table, with 10 rats in each group. The animal model of postoperative fatigue syndrome was simulated by the by small bowel resection and anastomosis. After 10 days of modeling, 1/2 reduced size orthotopic liver transplantation surgery was performed. After the surgery, the salidroside group was treated by tail intra-venous injection of salidroside for 7 days. The levels of peripheral blood 5-efferent neurotransmitter serotonin (5-HT), pituitary growth hormone (GH), cerebral cortex potentials and liver function of aspartate aminotransferase (AST), biliru-bin (TBIL), alanine aminotransferase (ALT) of the three groups were compared. Results The levels of 5-HT, AST, TBIL and ALT in the model group after 7 days of treatment were respectively (9.93 ± 0.22) ng/mL, (504.7 ± 37.9) U/L, (54.87 ± 3.21)μmol/L, (181.6 ± 18.3) U/L, which were significantly higher than (16.23 ± 0.52) ng/mL, (554.3 ± 32.9) U/L, (76.87±4.22)μmol/L and (312.3±34.5) U/L after 10 days of modeling;while the level of GH in the model group after 7 days of treatment was (0.71± 0.03) pg/mL, which was significantly lower than (0.53± 0.05) pg/mL after 10 days of mod-eling;the cerebral cortex discharge increased gradually from (459.23± 48.08) mV after 10 days of modeling to (513.03 ± 50.28) mV after 7 days of treatment;Compared with the model group before modeling and the sham group, the differences were statistically significant (P<0.05). Compared with the sham group, the levels of 5-HT, AST, TBIL and ALT in the sali-droside group after 10 days of modeling were respectively (10.72 ± 0.31) ng/mL, (484.7 ± 20.1) U/L, (55.21 ± 5.21) mol/L, (186.2±19.2) U/L, which increased significantly (P<0.05);GH was (0.75±0.02) pg/ml, which decreased significantly (P<0.05); the cerebral cortex discharge was (451.71 ± 45.58) mV, which gradually increased (P<0.05). Compared with the salidroside group after 10 days of modeling and the model group, the levels of 5-HT, cerebral cortex discharge, AST of liver function, TBIL and ALT in the salidroside group after 7 days of treatment were respectively (10.21 ± 0.31) ng/mL, (321.12±38.24) mV, (194.3±22.1) U/L, (24.07±1.25) mol/L, (112.2±23.1) U/L, which decreased significantly (P<0.05); GH was (2.67 ± 0.11) pg/mL, which increased obviously (P<0.05). Conclusion Salidroside can reduce fatigue syn-drome after liver transplantation in rabbits.
