| 摘 要： | 目的体外实验初步探讨叶下珠醇提物对人胃癌细胞MKN28生长的影响。方法通过集落形成试验观察叶下珠醇提物对人胃癌细胞MKN28集落形成的影响;将不同浓度叶下珠醇提物作用于MKN28细胞,倒置显微镜下观察细胞形态变化,MTT法测定叶下珠醇提物对MKN28细胞的增殖抑制率;流式细胞仪检测叶下珠醇提物作用48 h后MKN28细胞的凋亡情况。结果在2 mg/mL和1 mg/mL 2个浓度下叶下珠醇提物对MKN28胃癌细胞集落形成具有不同程度的抑制作用,浓度大者,抑制作用更强。MTT法测定细胞增殖抑制率结果表明,叶下珠醇提物对MKN28细胞增殖有抑制作用,其抑制效应具有浓度和时间依赖性。流式细胞检测提示叶下珠醇提物能诱导人胃癌细胞MKN28发生凋亡。结论叶下珠醇提物能抑制人胃癌细胞MKN28增殖,凋亡是其作用机制之一。
