非霍奇金淋巴瘤患者乳腺癌耐药蛋白的表达及其与药物敏感性的关系

摘    要:目的 探讨非霍奇金淋巴瘤(NHL)患者乳腺癌耐药蛋白(BCRP)的表达及其与药物敏感性的关系.方法 采用RT-PCR法检测比例NHL和5例慢性淋巴结炎患者新鲜淋巴组织标本中BCRP及多药耐药基因(MDR1)的表达水平;并对所有患者进行随访.结果 5例慢性淋巴结炎患者BCRP和MDR1表达均阴性;66例NHL患者BCRP阳性率45.5%,MDR1阳性率59.1%;T细胞淋巴瘤患者BCRP阳性率高于B细胞淋巴瘤者,NHL Ⅲ~Ⅳ期患者BCRP阳性率高于Ⅰ~Ⅱ期者(均P〈0.05);BCRP阳性患者的复发率高于阴性者,而缓解率和5年生存率均低于阴性者(均P〈0.05);BCRP和MDR1阳性、BCRP阳性和MDR1阴性、BCRP阴性和MDR1阳性、BCRP和MDR1阴性患者的化疗疗效均不同(均P〈0.05).结论 本组NHL患者BCRP和MDR1表达率较高,可能为化疗敏感性差的重要原因.

Relationship between expression of breast cancer resistance protein (BCRP) and drug sensitivity in patients with non-Hodgkin's lymphoma

Abstract:Objective To investigate the relationship between the expression of breast cancer resistance protein (BCRP) gene and drug resistance in patients with non-Hodgkin's lymphoma (NHL). Methods The expression of BCRP and multidrug resistance (MDR1) genes in and 66 NHL cases and 5 cases of chronic lymphadenitis The expression of BCRP and MDR1 genes were negative in 5 cases of chronic lymphadenitis; while the positive rate of BCRP gene was 45.5% and MDR1 gene was 59.1% in 66 NHL cases. The expression of BCRP gene was associated with pathologic types and clinical stages of NHL patients. The expression of BCRP gene was also associated with remission rate, the recurrence rate and the 5-year survival rate of patients. There were different response rate to treatment among the NHL patients with BCRP positive MDR1 positive, BCRP positive MDR1 negative, BCRP negative MDR1 positive or BCRP negative MDR1 negative (P〈0.05). Results The expression of BCRP and MDR1 gene of 5 chronic lymphadenitis were negative; The positive ratio of BCRP gene was 45.5% and MDR1 gene was 59.1% respectively in 66 NHL. There was different positive ratio of BCRP gene in different pathologic type and clinical stages patients. The remission rate, the recurrence rate and the 5 years survival rate were related with the expression of BCRP gene ; The response of treatment were different among the group of BCRP positive MDR1 positive,BCRP positive MDR1 negative,BCRP negative MDR1 positive,BCRP negative MDR1 negative (P〈0.05). Conclusion The poor response to therapy is closely related to the high expression of BCRP and MDR1 genes in patients with non-Hodgkin's lymphoma.
