摘    要:目的观察丁烯酸内酯(BUT)对软骨细胞的毒理作用以及维生素C(VC)、维生素E(VE)和硒(Se)的保护作用。方法体外培养软骨细胞,加入不同浓度BUT以及VC、VE和Se。用MTT法检测细胞活性,收集培养液检测氧化相关指标丙二醛(MDA)、谷胱甘肽(GSH)、超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-PX)的表达水平,通过光镜和电镜来观察其形态变化。结果BUT可以导致软骨细胞膜系统受损甚至死亡。微量毒素可以刺激软骨细胞的活性,增强其抗氧化作用;大量毒素则表现为抑制作用。VC、VE和Se对这种损伤的保护作用很有限。结论BUT通过刺激细胞产生内源性的氧自由基来发挥其损伤作用,进而导致细胞膜系统破坏甚至细胞死亡;单一的抗氧化剂并不能完全拮抗这种损伤作用。

Abstract:Objective To observe the toxic effect of Butenolide (BUT) on chondrocytes and the protection of vitamin C (VC), vitamin E (VE) and selenium (Se). Methods Chondrocytes were cultured in vitro with different concentration of BUT, and with/without VC, VE or Se. The cell activity was detected by MTT, and the catalytic activity of SOD, GSH-PX and content of MDA, GSH in culture media were measured. Optical microscope and electron microscope were used to observe the morphological changes. Results BUT damaged chondrocytes especially the membrane system. With the increasing concentration of BUT the damage of chondrocytes was increased first then decreased. After adding VC, VE and Se, the damage caused by BUT was not relieved. Conclusion BUT can cause the damage of membrane system resulting the death of chotrocytes; VC, VE and selenium do not show protective effects.
