白花蛇舌草与半枝莲复合提取物对膀胱癌YTS-1细胞系抑制作用的研究

摘    要:目的探讨白花蛇舌草与半枝莲复合提取物对膀胱癌YTS-1细胞系的作用。方法常规方法培养膀胱癌YTS-1细胞株,取对数生长期细胞进行实验。采用MTT法观察不同浓度药物(0.25、0.5、1、2、4和8mg/ml)对细胞的生长抑制率;流式细胞技术观察2mg/ml药物作用24、48、72h后细胞周期的改变;克隆形成实验检测不同浓度药物(0.25、0.5、1.0、2.0、4.0mg/m1)处理后细胞克隆形成率的变化。结果(1)不同浓度白花蛇舌草与半枝莲复合提取物作用于YTS-1细胞系24、48、72h后,在0.25-2mg/ml范围内随着药物浓度的增加和作用时间的延长,其对YTS-1细胞的生长抑制率也相应升高,抑制作用表现出良好的量效和时效依赖关系(P〈0.05)。(2)实验组2mg/ml白花蛇舌草与半枝莲复合提取物作用48h后,G2/M期细胞明显减少(P〈0.05);作用72h后G2/M期细胞显著减少(P〈0.01)。在sub-G1区域出现了一个细胞峰,表明YTS-1出现了细胞凋亡迹象。(3)空白对照及025、0.5、1.0、20、40mg/ml的白花蛇舌草和半支莲复合提取物作用YTS-1细胞后克隆数分别为216.6±18.50、156.7±16.29、81.33±6.66、43.00±6.56、37.67±14.22、43.00±12.12,克隆形成率分别为10.8%、0.78%、0.41%、0.21%、0.18%、0.22%,除0.25mg/ml浓度处理组与空白对照组克隆形成率无明显差异外(P〉0.05),其余各浓度处理组克隆形成率均显著低于空白对照组(P〈0.01)。结论白花蛇舌草和半支莲复合提取物可以抑制膀胱癌YTS-1细胞集落的形成,诱导细胞凋亡,具有明显的肿瘤抑制作用。

Inhibitive effects of compound of Oldenlandia and Scutellaria Barbata D. Don on human bladder cancer cell line YTS-1 in vitro

Abstract:Objective To investigate the inhibitive effects of compound of Oldenlandia and Scutellaria Barbata D. Don on human bladder cancer cell line YTS-1 in vitro. Methods The cultured YTS-1 cells were treated with compound of Oldenlandia and Scutellaria Barbata D Don (0.25, 0.5, 1, 2, 4 and 8mg/ml) . Cell growth inhibition rate was determined with MTT assay; cell-cycle was detected by flow cytometry at 24, 48, 72 h after cells were treated with 2mg/ml compound. Clones formation rate of YTS-1 was also assayed after cells were exposed to different concentrations of drugs (0.25, 0.5, 1, 2, 4 and 8mg/ml). Results The growth of YTS-1 cells were inhibited by different concentrationc of compound in a dose and time-dependent manner, at the concentration range of 0.25-2mg/ml and time range of 24-72h (P〈0.05). Flow cytometry showed reduced proportion of G2/M cell arrest (P〈 0.05) and the hypodiploid DNA peak appeared at 72h after treatment of 2mg/ml of the compound. After treatment of compound at 0.5mg/ml, 1mg/ml, 2mg/ml, 4mg/ml the colony formation rates were 0.78%, 0.41%, 0.21% and 0.18% respectively, that was 0.22% in control group (P〈0.05). Conclusion Compound of Oldenlandia and Scutellaria Barbata D. Don can inhibit bladder cancer cells through inducing cell apoptosis and reducing colony formation in vitro.
