多穗柯提取物长期毒性研究

摘    要:目的观察多穗柯提取物重复灌胃对大鼠产生的毒性反应。方法高剂量组(2g提取物·kg-1·d-1)、中剂量组(1g提取物·kg-1·d-1)、低剂量组(0.5g提取物·kg-1·d-1)和空白对照组,大鼠各50只,灌服多穗柯提取物,连续25周,分别于给药中期、停药时和停药后2周,取部分大鼠进行16项血液学指标、15项血清生化学指标检查,显微观察29处组织病理变化。结果各给药组在给药期间血糖明显低于对照组(P〈0.05),高剂量组停药时肝功能指标显著高于对照组(P〈0.05),且肝细胞变性,其余各组检查未见明显异常。结论多穗柯提取物经口给药有降低大鼠血糖作用,长期大剂量给药可见可逆性肝中毒改变外,未见明显的其他毒性反应。

Abstract:Objective To study the long-term toxicity of the extract of Lithocarpus litseifolius in rats by repeated oral administration.Methods Daily doses of the extract at 2.0 g/kg,1.0 g/kg,and 0.5 g/kg for 25 weeks were used for high,mid,and low dose group(50 rats each),respectively.Examinations including 16 haematologicalindexes,15 biochemical indexes,and 29 organs(histopathology) were performed at interim administration,withdrawal and 2 weeks after the time,respectively.Results During the administration of glucose in treated group was significantly lower than that in control group(P 0.05) .The liver function indexes in high dose group were significantly higher than those in control group(AST and ALT activities),and their liver cells degenerated.No changes were found in other dose groups.Conclusion Oral administration of the extract of Lithocarpus litseifolius could lower the blood glucose level.Repeated administration of this extract could lead to reversible damage to the liver function.
