通窍活血汤水提取物对重型颅脑损伤大鼠血清白介素-6、白介素-10水平的影响

目的 探讨通窍活血汤水提取物对重型颅脑损伤大鼠血清炎症因子白介素(IL)-6、IL-10的影响.方法 将72只大鼠随机分成正常对照组、模型组及通窍活血汤组,每组24只.制备重型颅脑损伤大鼠模型,造模后6h通窍活血汤组给予通窍活血汤水提取物灌胃,正常对照组、模型组给予生理盐水灌胃.将正常对照组、模型组、通窍活血汤组每组中的24只大鼠各随机分配到3个时间处理组,分别于造模后24h、72 h、168 h采血并处死,检测其血清IL-6、IL-10水平,比较各组大鼠在不同时点上IL-6、IL-10的表达差异.结果 (1)3组间IL-6、IL-10水平比较差异有统计学意义,IL-6、IL-10水平均有随时间变化的趋势,分组与时间有交互效应(P均＜0.05).(2)模型组、通窍活血汤组各时点IL-6水平均高于正常对照组,且均在24 h处于峰值,并分别于168 h、72 h降至最低值;通窍活血汤组IL-6水平在24h、72 h两个时点低于模型组(P均＜0.05).(3)模型组、通窍活血汤组各时点IL-10水平均高于正常对照组,通窍活血汤组168 h时点IL-10水平高于其24 h、72 h时点(P均＜0.05).结论 通窍活血汤可以减轻重型颅脑损伤继发的炎症反应.

Effect of water extract of Tongqiao Huoxue decoction on serum levels of interleukin-6,interleukin-10 in rats with severe brain injury

Objective To explore the effect of water extract of Tongqiao Huoxue decoction on the levels of serum inflammatory factor interleukin(IL)-6,IL-10 in rats with severe brain injury.Methods A total of 72 rats were randomly divided into normal control group,model group and Tongqiao Huoxue decoction group,with 24 rats in each group.After 6 hours of severe brain injury rat model's successful development,the Tongqiao Huoxue decoction group was treated with the water extract of Tongqiao Huoxue decoction by gavage,while the normal control group and the model group were given normal saline by gavage.Twenty-four rats in each group of the normal control group,the model group and the Tongqiao Huoxue decoction group were randomly assigned to 3 time treatment groups separately and the blood samples were collected at 24,72 and 168 hours after modeling,then the rats were executed and the levels of serum IL-6 and IL-10 were detected.The expressions of IL-6 and IL-10 of rats at diverse time-points and in different groups were compared.Results ① The levels of serum IL-6 and IL-10 among the 3 groups were statistically significant and had a tendency to change with time,and there was an interactive effect between group and time (all P ＜ 0.05).② The levels of IL-6 in the model group and Tongqiao Huoxue decoction group at each time-point were higher than those in the normal control group,and reached peak value on the 24th hour,then decreased to the lowest value on the 168th hour,72nd hour respectively;the levels of IL-6 in the Tongqiao Huoxue decoction group were lower than the levels in the model group on the 24th and 72nd hour(all P ＜0.05).③ The levels of IL-10 in the model group and Tongqiao Huoxue decoction group at each time-point were higher than those in the normal control group,and the level of IL-10 on the 168th hour was higher than the level on the 24th or 72nd hour in the Tongqiao Huoxue decoction group (all P ＜ 0.05).Conclusion Tongqiao Huoxue decoction can relieve the inflammatory response secondary to severe traumatic brain injury.
