摘    要:目的探讨鞘氨醇-1磷酸盐(S1P)对宫颈癌HeLa细胞生长和侵袭能力的影响。方法分别应用S1P(实验组)和无脂肪型牛血清白蛋白(BSA)载体溶液(对照组)处理宫颈癌HeLa细胞,采用噻唑蓝和5-溴脱氧尿嘧啶核苷标记法细胞增殖分析法检测肿瘤细胞活力和增殖变化,趋化小室和侵袭小室检测肿瘤细胞迁移和侵袭能力变化。结果与对照组细胞相比,实验组HeLa细胞活力及细胞增殖、HeLa细胞迁移和侵袭能力显著增强(P〈0.05)。结论 S1P可以导致宫颈癌HeLa细胞生长和侵袭能力增强,提示S1P蛋白在宫颈癌进展中可能发挥重要作用。

Abstract:Objective To explore the effect of sphingosine-1 phosphate( S1P) on cell growth and invasion in cervical cancer cell line HeLa. Methods Cervical cancer HeLa cells were treated with S1P( experimental group) or fatty acid-free bovine serum albumin( BSA)( control group),MTT and Brd U cell proliferation assays were used to analyze cell viability and proliferation. The cell migration and invasion was detected using Boyden chamber and Transwell assays. Results As to HeLa cells,the cell viability and proliferation as well as cell migration and invasion in the experimental group increased significantly compared with control group( P〈0. 05). Conclusion S1 P can lead to the enhancement of cell growth and invasion in cervical cancer HeLa cells,which suggests that S1 P protein may play a key role in the progression of cervical cancer.
