摘    要:目的:在体外探讨α-干扰素(IFN-α)对人脐血来源的树突状细胞(DC)分化成熟及功能的影响。方法:取lO份脐血分离单个核细胞,常规DC诱导培养条件中加入不同剂量的IFN—α(依剂量0、100、500U/m1分为A、B、C3组)。流式细胞仪分析CD83等细胞表面标志,ELISA检测IL-12等细胞因子的表达,MTT法检测DC对同种异体T淋巴细胞的增殖能力。结亲:在常规DC诱导培养条件中加入IFN—α培养的贴壁单个核细胞,在第5天即可见B、C组细胞表面呈树突状突起.第7天B、C组诱导的细胞其DC特异性表面标志CD83、HLA—DR、CD86较A组明显升高,且随IFN-α剂量增加而增加(P〈O.05);B、C组分泌的细胞因子IL-12、IFN-γ较A组均增高,而IL-10较A组减少(P〈O.05);B组和C组DC刺激T淋巴细胞增殖的能力显著高于A组(P〈O.05),又以T与DC比为10:1时最明显。结论:在常规DC诱导培养基础上.再加入IFN-α能进一步促进脐血单个核细胞向具有DC典型形态及更强功能的成熟DC转化。

Abstract:Objective: To study the influence of interferon-α on differentiation, maturation and im- mune function of human cord blood-derived dendritic cells in vitro. Methods: Mononuclear cells were iso- lated from ten samples of cord blood and each sample was divided into 3 groups (A, B and C). In conven- tional DC induction condition, different doses (0,100 and 500 U/ml) of IFN-α were added into A,B,C groups, respectively. Cell morphous were observed by microscope ,cell surface markers such as CD83 were analysis by FACS, cytokines such as IL-12 were detected by ELISA, and the capability of DC stimulating allogeneic T cell proliferation was assayed by MTF. Results: After 5 days, typical DC morpholology could be seen in B and C groups. On 7th day, surface marker CD83, HLA-DR and CD86 expressed higher in B and C groups than those in A group (P〈0.05). IL-12 and IFN-γ secreted by DC in B and C groups were higher than those in A group, while IL-IO lower in B and C groups than those in A group (P〈0.05). The capability of DC stimulating T cell proliferation in B and C groups was higher than that in A group (P〈0.05), espe- cially when the ratio of DC to T cell was 1:10. Conclusion: The findings suggest that, in conventional DC induction condition, adding IFN-α could promote cord blood-derived mononuclear cells to convert into mature DC with typical morphous and stronger function.
