大黄zhe虫丸血浆药理学与血清药理学作用的比较研究

摘    要:目的建立血浆药理学研究方法,探讨话血化瘀方剂大黄(庶虫)虫丸对血小板聚集的影响.方法健康志愿者54人,其中5人服大黄(庶虫)虫胶囊,5人服阿司匹林用于制备含药的血清和血浆,44人不服药,采血后分离血小板PRP,随机分成8组自体血清组、异体血清组、大黄(庶虫)虫血清组、阿司匹林血清组、自体血浆组、异体血浆组、大黄(庶虫)虫血浆组、阿司匹林血浆组.每组分别加入相应的血清和血浆孵育30 min,用ADP和肾上腺素二种诱导剂处理,观察各组对血小板聚集的影响.结果用ADP和肾上腺素二种诱导剂处理,血小板在自体血清组、异体血清组、大黄(庶虫)虫血清组、阿司匹林血清组中均不聚集,而在自体血浆组、异体血浆组、大黄(庶虫)虫血浆组、阿司匹林血浆组中聚集.用ADP诱导自体血浆组与异体血浆组相比较,P1(1 min聚集率)、P5(5 min聚集率)、PM(最大聚集率)、TM(解聚集率)、I(聚集时间)差异均无显著性(P>0.05).大黄(庶虫)虫胶囊和阿斯匹林组与异体血浆组相比较,P1、P5、PM、I差异均有非常显著性(P<0.01),TM也显著下降(P<0.05).大黄(庶虫)虫胶囊与阿斯匹林组比较,P1、PM、I差异均有显著性(P<0.05),而P5、TM二者作用相似.用肾上腺素诱导自体血浆组与异体血浆组相比较,P1、P5、PM、TM、I差异均无显著性(P>0.05).大黄(庶虫)虫胶囊和阿斯匹林组与异体血浆组相比较,P5、PM、I差异均有非常显著性(P<0.01),而P1、TM差异也有显著性(P<0.05).大黄(庶虫)虫胶囊与阿匹林组比较,P1、P5、PM差异均有显著性(P<0.05),而TM、I二者作用相似.结论在进行血栓与止血体外研究时,用血清药理学方法是无法完成的.血浆药理学方法能更好地再现体内环境中产生的药理效应.大黄(庶虫)虫丸抗血小板聚集作用明显优于阿司匹林.

Abstract:Objective To develop a plasma pharmacological method for evaluating the effect of dahuang zhechong capsule on platelet aggregation which is a representative traditional Chinese medicine patent presciption promoting blood circulation by removing blood stasis. Methods Platelets specimens from 54 healthy volunteers,5 of them took dahuang zhechong capsule,5 of them took aspirin,then serum and plasm with medicine would be made ,44 didn' t take medicine, platelet PRP were separated after with drawing blood, which were divided into 8 groups,i. e. auto-serum, allo-serum, serum with dahuang zhechong capsule, serum with aspirin, auto-plasma, allo-plasma, plasma with dahuang zhechong capsule, plasma of aspirin. After ADP and adrenalin were added into the specimens and hatched for 30 min, the effects of specimens of platelet aggregation were observed. Results After ADP and adrenalin were added,all the serum groups did not present platelet aggregation, whose images showed a straight line, while all the plasma groups presented platelet aggregation. P1 ,P5 ,PM,I,and TM have no significant differences (P >0. 05) between auto-plasma group and alloplasma group induced by ADP. P1 ,P5 ,I and PM have significant differences (P <0. 01) and TM decreased significantly (P<0.05) comparing plasma group with dahuang zhechong capsule and plasma group of aspirin to allo-plasma group. P1 ,PM and I have significant differences (P <0. 05) ,and P5 and TM are simulate comparing plasma group with dahuang zhechong capsule to plasma group of aspirin. P1 ,P5 ,PM ,TM and I have no significant differences (P > 0. 05) comaring auto-plasma group to allo-plasma group induced by adrenalin. P5,PM and I have significant differences,and P1 and TM have sinificant differences (P <0. 05) comparing plasma group with dahuang zhechong capsule and plasma group of aspirin to allo-plasma group. P1 ,P5 and PM have significant differences ( P < 0. 05 ) , and TM and I are simulate comparing plasma group with dahuang zhechong capsule to plasma group of aspirin. Conclusion The serum pharmacological study is inappropriate to study platelet aggregation in vitro. The plasma pharmacological study can well reflect the pharmacological effect prduced in vivo. Dahuang zhechong capsule has better effect of anti artery thrombosis than that of aspirin.
