保妇康栓对人乳头状瘤病毒抑制作用的实验研究

摘    要:目的:探讨中药制剂保妇康栓对人乳头状瘤病毒(HPV)16亚型的抑制作用。方法:以不同浓度的含保妇康栓培养基体外培养宫颈癌细胞系CaSki和宫颈永生化细胞H8,相差显微镜下观察活细胞形态学变化;采用四甲基偶氮唑蓝比色法以及流式细胞术技术检测不同浓度保妇康栓对细胞增殖的影响;应用逆转录-聚合酶链反应技术(RT-PCR)检测人乳头状瘤病毒HPV16亚型E6E7的表达。结果:不同浓度的保妇康栓可以抑制细胞的增殖;在药物作用下,CaSki细胞G1期细胞增加(P<0.05),G2、S期细胞减少(P<0.05),CaSki细胞凋亡率高于对照组(P<0.05),而对H8细胞周期和凋亡率影响不明显;两种细胞HPV16E6E7基因片段mRNA表达均明显低于对照组(P<0.01)。结论:保妇康栓在体外抑制宫颈癌细胞系CaSki和宫颈永生化细胞H8增殖,其机制可能是通过抑制HPV16E6E7表达而抑制肿瘤细胞生长。

Abstract:Objective:To investigate the inhibition of human papillomavirus(HPV)16 by Chinese patent medicine-Baofukang suppository.Methods:The human cervical cancer cell line-CaSki and the immortalized cervical epithelial cell-H8 were cultured with different concentration of Baofukang suppository.Changes of cellular form were observed by phase contrast microscope.The proliferative inhibition of different concentration of Baofukang suppository on CaSki and H8 was measured by methyl thiazolyl tetrazolium(MTT)assay and flow cytometry assay.The mRNA expression of HPV16E6E7 was determined by RT-PCR semi-quantitatively.Results:The growth of CaSki and H8 was inhibited by different concentration of Baofukang suppository.CaSki cells were accumulated in G1 phase,while in G2 and S phase,the cells decreased(P<0.05).The apoptosis rate in CaSki cells was higher than that in control group.But for H8 cells,there was no significant change on cell cycles or on the rate of apoptosis.After Baofukang suppository treatment,CaSki and H8 cell line showed decreased level of HPV16E6E7 mRNA than that in the control group.Conclusion:Baofukang suppository may execute its antiHPV activity primarily by decreasing the expression of HPV16E6E7 in cervical cancer cell line and inhibit the growth of cervical epithelial cell.
