成骨生长肽对大鼠骨髓基质细胞增殖分化的影响

摘    要:目的 研究成骨生长肽(osteogenic growth peptide, OGP)在体外对骨髓基质细胞增殖和成骨向分化的影响.方法 在体外培养的大鼠骨髓基质细胞中加入不同浓度的OGP(10-11~10-7 mol/L),用MTT法检测细胞增殖,酶联免疫法检测细胞内碱性磷酸酶(ALP)的表达,RT-PCR法检测细胞内核心结合因子1(Cbfa1) mRNA的表达.结果 OGP对骨髓基质细胞增殖有促进作用(P《0.05),最大效应浓度为10-9 mol/L;能增加细胞内ALP活性(P《0.05),最大效应浓度为10-8 mol/L;可诱导Cbfa1 mRNA的表达(P《0.05).结论 OGP可促进骨髓基质细胞的增殖,并通过诱导其内Cbfa1 mRNA的表达促进其成骨向的分化.

Abstract:Objective To elucidate the effect of osteogenic growth peptide(OGP) on bone marrow stromal cells proliferation and differentiation in vitro. Methods Bone marrow stromal cells were cultured individually in medium contained various concentrations of OGP; and cell proliferation quantity, alkaline phosphatase activity and level of Cbfa1 mRNA were examined by means of MTT, enzyme-linked immunoreaction and reverse transfer polymerase chain reaction respectively. Results In vitro, the addition of OGP in cultures resulted in an increase of cell proliferation(P<0.05), ALP activity(P<0.05) and level of mRNA for Cbfa1(P<0.05). All concentrations (10-11- 10-7 mol/L) of OGP were effective but the maximum response was obtained with 10-8 mol/L OGP. Conclusion Our results demonstrate that OGP stimulates proliferation and sequential differentiation of bone marrow stromal cells in vitro.
