摘    要:目的研究重组生长激素(recombinant growth hormone,rGH)对高胆固醇饮食的新西兰兔的血脂调节作用及rGH对肝脏低密度脂蛋白(low density lipoprotein,LDL)受体mRNA表达的影响.初步探讨rGH影响血脂的机制.方法①健康新西兰白兔,均为雄性,兔龄4个月,体质量(2.2±0.3)kg,随机分组观察高胆固醇饮食的条件下,rGH对血脂的影响.②RT-PCR技术测定兔肝低密度脂蛋白受体(low density lipoprorein receptor,LDL-R)mRNA的表达情况.结果①rGH组血清总胆固醇(total cholesterol,TC)、低密度脂蛋白胆固醇(low density lipoprotein cholesterol,LDL-C)低于对照组(P<0.01)但对甘油三酯(TG)无明显影响.②rGH组LDL-RmRNA的表达高于对照组(P<0.01).结论rGH能够有效的降低高胆固醇饮食条件下的TC、LDL-C,2.rGH降血脂的作用,可能与rGH促进LDL-R mRNA表达上调有关.

Abstract:Objective To study the effects of recombinant growth hormone (rGH) on serum lipid levels and the expression of hepatic LDL receptor mRNA of rabbits by high-cholesterol diet. Methods Four-month-old male rabbits with high-cholesterol diet were randomly divided into two groups: control group and rGH group. Serum lipid levels were measured after 1 or 3 months. Expression of LDL receptor mRNA in liver was determined by RT-PCR. Results As compared with control group, rGH group had lower levels of serum TC, LDL-C, higher levels of LDL receptor mRNA in liver, but no change for serum TG. Conclusion rGH enhanced the expression of LDL receptor mRNA in liver, which may be related to the mechanisms of rGH lowering serum lipid levels.
