苦参碱及氧化苦参碱抑制A549细胞增殖及诱导细胞凋亡的比较研究

目的观察比较苦参碱及氧化苦参碱对A549细胞系增殖能力的影响及其对A549细胞凋亡的诱导效应.方法以不同剂量的苦参碱及氧化苦参碱分别作用A549细胞,观察细胞形态,MTT法检测细胞的增殖能力,TNUEL法原位检测DNA断裂,流式细胞仪检测凋亡.结果苦参碱在浓度为0.5g/L时即表现出明显的抑制A549增殖的作用,浓度在1.0g/L时TUNEL原位检测DNA断裂为强阳性,流式细胞仪检测出凋亡峰;相同浓度的氧化苦参碱没有明显的抑制A549增殖的作用,当其浓度增加到10倍出现对A549抑制增殖的作用,但是TUNEL原位检测DNA断裂为弱阳性,流式细胞仪不能检测出凋亡峰.结论苦参碱可抑制A549细胞系增殖并诱导其凋亡;相同浓度下氧化苦参碱不能抑制A549细胞系增殖,10倍剂量也未出现细胞凋亡.

Effects of matrine and oxymatrine on the proliferation and the apoptosis of A549 cells

Objective To observe the effects of matrine and oxymatrine on the proliferation and the apoptosis of A549 cells. Methods The morphological changes of A549 cells after treatment with different concentrations of matrine and oxymatrine were observed. The proliferative ability, the DNA breakage, and the apoptosis of A549 cells were detected by MTT method, in situ TUNEL method, and flow cytometry, respectively. Results The proliferation of A549 cells was remarkably inhibited by 0.5 g/L matrine. Strong positive DNA breakage was detected by TUNEL and the apoptotic peak was detected by flow cytometry in A549 cells after treatment with 1.0 g/L matrine. Nevertheless, oxymatrine at the same concentrations had no obvious inhibitory effect on the proliferation of A549 cells. When the concentration of oxymatrine was increased as high as 10 times, the proliferation of A549 cell was inhibited, and weakly positive DNA breakage was detected, but the apoptotic peak was not detected. Conclusion Matrine can inhibit the proliferation and induce apoptosis of A549 cells at some concentration, but oxymatrine at the same concentration has no inhibitory effect on the cell proliferation, and can not induce the cell apoptosis even at the concentration as 10 times as that of matrine.
